The assembly of Tcrb and Tcra genes require double negative (DN) thymocytes to undergo multiple rounds of programmed DNA double-strand breaks (DSBs), followed by their efficient repair. However, mechanisms governing cell cycle checkpoints and specific survival pathways during the repair process remain unclear. Here, we report high-resolution scRNA-seq analyses of individually sorted mouse DN3 and DN4 thymocytes, which reveals a G2M cell cycle checkpoint, in addition to the known G1 checkpoint, during Tcrb and Tcra recombination.

The use of 3D-printed electrodes is reported fabricated from in-house conductive filament composed of a mixture of recycled poly (lactic acid) (rPLA), graphite (Gpt), and carbon black (CB) for fast detection of the abused drug ketamine. This study focused on evaluating the fractionation of pequi oil and modeling the process using solid-liquid equilibrium (SLE) theory. The pequi oil was comprehensively characterized, including its fatty acid (FA) and acylglycerol (AG) profiles, moisture content, acidity, carotenoid levels, and thermal behavior. Low acidity and partial acylglycerols content, along with its TAG profile (mainly OOP, POP, OOO and PPP) and melting behavior proved that, in fact, this oil is quite suitable for fractionation.

This study aimed to investigate the impact of sucrose reduction on the sensory dynamics and consumer acceptance of short-dough biscuits, incorporating thaumatin as a flavor enhancer. Using the hedonic threshold, the research identified that a 25 % reduction in sucrose was generally acceptable to consumers, whereas a 50 % reduction led to product rejection, indicating a strong preference for sweetness. Temporal Dominance of Sensations (TDS) analysis further confirmed consumers' sensitivity to sugar levels, with a marked preference for biscuits containing a 25 % sucrose reduction.
