Three-dimensional (3D) cardiomyocyte spheroids are essential models to replicate cardiac structural and functional features in vitro. However, conventional planar and rigid microelectrode arrays (MEAs) suffer from low-quality electrophysiological recording of 3D cultures, due to limited contact areas and weak coupling between cells and MEA chips. Herein, we developed a PEDOT: PSS-modified organic flexible and implantable MEA (OFI-MEA) coupled with a self-developed integrated biosensing platform to achieve high-throughput, long-term, and stable bidirectional internal electrophysiology in 3D cardiomyocyte spheroids.

Williams syndrome (WS; OMIM#194050) is a rare disorder, which is caused by the microdeletion of one copy of 25-27 genes, and WS patients display diverse neuronal deficits. Although remarkable progresses have been achieved, the mechanisms for these distinct deficits are still largely unknown. Here, we have shown that neural progenitor cells (NPCs) in WS forebrain organoids display abnormal proliferation and differentiation capabilities, and synapse formation.

The spontaneous emergence of lipid vesicles in the absence of evolved biological machinery represents a major challenge for bottom-up synthetic biology. We show that coacervate microdroplets could create a compartmentalized environment that enriches lipid molecules and facilitates their spontaneous assembly into lipid vesicles. These vesicles can escape from the coacervate microdroplets in a continuous process under non-equilibrium conditions, resembling a constant production process akin to a "primitive enzyme" factory assembly line.

Microscopy is an indispensable tool for collecting biomedical information in pathological diagnosis, but manual annotation, measurement and interpretation are labor-intensive and costly. Here, we propose a task-driven framework powered by large models that excel in visual analysis and real-time control, paving the way for the next generation of microscopes. We achieve proof-of-concept success on clinical tasks, specifically in adaptive analysis of H&E-stained liver tissue slides.

The leucine zipper-like transcriptional regulator 1 (LZTR1) gene has been reported to be associated with many kinds of human diseases, including cardiac disease, Noonan syndrome, and schwannomatosis. In this study, peripheral blood mononuclear cells (PBMCs) derived from patient diagnosed with dilated cardiomyopathy (DCM) was successfully reprogrammed into the human induced pluripotent stem cells (iPSCs) line, harboring a distinct heterozygous mutation in the LZTR1 gene. The established patient-derived iPSCs expressed endogenous pluripotent markers, demonstrated the potential to differentiate into three germ layers (endoderm, mesoderm, and ectoderm), and exhibited a normal karyotype.

Biofilm-associated surgical site infection (BSSI) is a common and grievous postoperative complication lacking effective remedies, mainly due to the poor drug accumulation and penetration in the biofilms featured by dense extracellular polymeric substances (EPSs). Here, it is found that the vascular cell adhesion molecule-1 (VCAM1) is highly overexpressed in the vascular cells of BSSI. It is proposed that the combination of VCAM1-mediated transcytosis and ultrasonic cavitation can consecutively overcome the biological barriers of vascular endothelial cells and EPS for biofilm eradication.

Hydrogen has been widely used in industrial and commercial applications as a carbon-free, efficient energy source. Due to the high flammability and explosion risk of hydrogen-air mixtures, it is vital to develop sensors featuring fast-responding and high sensitivity for hydrogen leakage detection. This paper presents a miniaturized electrochemical gas sensor by elaborately establishing a nanocomposite and thin ionic liquid interface for highly sensitive and rapid electrochemical detection of hydrogen, in which a remarkable response time and recovery time of approximately 6 s was achieved at room temperature.

Multidrug resistance (MDR) has emerged as a major barrier to effective breast cancer treatment, contributing to high rates of chemotherapy failure and disease recurrence. There is thus a pressing need to overcome MDR and to facilitate the efficient and precise treatment of breast cancer in a targeted manner. In this study, endogenous functional lipid droplets (IR780@LDs-FeO/OA) were developed and used to effectively overcome the limited diffusion distance of reactive oxygen species owing to their amenability to cascade-targeted delivery, thereby facilitating precise and effective sonodynamic therapy (SDT) for MDR breast cancer.

Objectives: To establish superb microvascular imaging (SMI) based thyroid imaging reporting and data system (SMI TI-RADS) for risk stratification of malignancy in thyroid nodules.
Methods: In total, 471 patients, comprising 643 thyroid nodules, who received conventional ultrasound (US), SMI, and a final diagnosis were extensively analyzed. A qualitative assessment of US features of the nodules was performed followed by univariable and multivariable logistic regression analyses, leading to the construction of the SMI TI-RADS, which was further verified using internal and external validation cohorts.
