High-performance liquid chromatographic method for determination of voriconazole in pure and gel formulation.

PLoS One

December 2024

Department of Pharmaceutical Chemistry, Baqai Institute of Pharmaceutical Sciences, Baqai Medical University, Gadap Town, Karachi, Pakistan.

Article Synopsis
  • A reliable RP-HPLC method with UV detection was developed for the accurate measurement of Voriconazole in pure and gel formulations, using a 50:50 acetonitrile-water mobile phase and a flow rate of 1.0 ml/min.
  • The method demonstrated excellent linearity (correlation coefficient = 0.9999) and precision, with a percentage relative standard deviation (RSD) of less than 2.0.
  • Robustness was evaluated using variables like flow rate and pH; while flow rate significantly affected drug concentration, the method remained straightforward and suitable for daily quality control with a retention time of under 4 minutes.

Photodegradation of drug substances leads to the formation of known and unknown degradation products. These unknown degradation products interfere and give erroneous results because of absorption on analytical wavelengths. This interference could be eliminated using the correction of irrelevant absorbancies.

Background: Increased expression of MRP 1 in AML patients results in the efflux of drugs from the cells, preventing the patient from achieving remission or potentially leading to relapse. Several studies have demonstrated that early identification of ABC transporter may yield favorable outcomes.

Aims And Objectives: The objectives of the study were to investigate the correlation between MRP 1 gene expression and MRP 1 protein levels and the response to remission induction in AML patients.

Background: L-Ascorbic acid (AA) is a highly unstable compound, thus, limiting its use in pharmaceutical and cosmetic products, particularly at higher concentrations.

Objective: This study aimed to stabilize the highly sensitive molecule (AA) by encapsulating it in β- cyclodextrin nanosponges (β-CD NS) that can be used further in preparing cosmeceuticals products with higher AA concentrations and enhanced stability.

Methods: The NS has been synthesized by the melting method.

Article Synopsis
  • Herbal medicines have historically been significant in treating cardiovascular disorders, and this study focuses on the edible fruit of Grewia asiatica in relation to hypertension.
  • The study utilized isolated atrial preparations from guinea pigs and rat aortic rings, finding that the crude ethanolic extract (Ga.Cr) reduced heart contractions and caused vasodilation in a dose-dependent manner.
  • Results indicated that Ga.Cr may lower blood pressure by inhibiting calcium influx and utilizing nitric oxide pathways, suggesting its potential as a natural treatment for hypertension.

Department of Pharmacology, Baqai Institute of Pharmaceutical Sciences, Baqai Medical University, 51 Deh Tor, Gadap Road, Near Toll Plaza, Super Highway, Karachi, Pakistan.

Article Synopsis
  • Cedrus deodara root oil has been traditionally used in Ayurveda for treating peptic ulcers, and this study assesses its effectiveness against ethanol-induced ulcers in rats.
  • The research involved 120 Wistar rats divided into four groups, with comparisons made between a control group, a positive control, a standard drug (Omeprazole), and a treatment group receiving C. deodara root oil.
  • Results indicated that C. deodara root oil exhibited anti-ulcer effects without causing significant damage to liver and kidney tissues, maintaining normal histological structures in the examined organs.

This is the first study on the photolysis of carboxymethylflavin (CMF), an intermediate in the photolysis of riboflavin (RF). CMF is photodegraded by removal of side-chain to lumichrome (LC) in acid solution and to LC and lumiflavin (LF) in alkaline solution. It also undergoes alkaline hydrolysis to 1,2-dihydro-1-methyl-2-keto-3-quinoxaline carboxylic acid (KA) and 1,2,3,4-tetrahydro-1-methyl-2,3-dioxoquinoxaline (DQ) by cleavage of isoalloxazine ring.
