Microglial polarization and ferroptosis are important pathological features in Alzheimer's disease (AD). Ghrelin, a brain-gut hormone, has potential neuroprotective effects in AD. This study aimed to explore the potential mechanisms by which ghrelin regulates the progression of AD, as well as the crosstalk between microglial polarization and ferroptosis.

Background: To evaluate the prognostic effect of endovascular treatment (EVT) in patients with acute base artery occlusion (ABAO) within 24 h of onset, and analyze the factors related to prognosis.

Methods: A retrospective analysis was conducted on all ABAO patients who received EVT within 24 h of onset in the neurology department of Baotou Central Hospital in Inner Mongolia from May 2016 to October 2022. Good prognosis was defined as a Modified Rankin Scale (mRS) score of 0-3 and poor prognosis mRS score of 4-6 at 90 day follow-up, and factors related to prognosis were analyzed.

Background: Circulating microRNAs (miRNAs) are recently a rapidly increasing of interest as non-invasive biomarkers for diagnosis and prognosis of acute myocardial infarction (AMI). Previous studies revealed that several miRNAs exhibited the capacity for diagnosis and prognosis of AMI, the reasons why these circulating miRNAs are concerned as targets for investigation are quite cryptogenic, presumably due to the lack of clues provided by global surveillance at the transcriptome level, and the current data for some miRNAs are controversial and inconsistent among independent studies.

Methods: To comprehensively profiling the potential miRNAs for diagnosis and prognosis of AMI, we reported transcriptomes of circulating miRNAs in the plasma of 27 healthy controls, 64 AMI patients (37 STEMI and 27 NSTEMI) and 20 AMI patients who were subjected to reperfusion therapy.

Article Synopsis
  • This study investigates the feasibility and pulmonary effects of low-pressure laparoscopic cholecystectomy (LPLC) for patients with cardiopulmonary comorbidities compared to standard-pressure laparoscopic cholecystectomy (SPLC).
  • It includes a multicenter, randomized controlled trial involving 144 patients, who were assigned to either the LPLC or SPLC group, focusing on postoperative carbon dioxide levels and other surgical outcomes.
  • Results showed no significant difference in carbon dioxide levels or pulmonary function between the groups, indicating that LPLC is a safe and feasible option for these patients.

Objective To explore the mechanism of butylphthalide (NBP) in regulating microglia activation and inflammatory cytokine expression in the hippocampus of the mouse model of delayed encephalopathy after carbon monoxide poisoning (DEACMP). Methods Wild-type C57 adult mice with normal cognitive function were selected, and DEACMP was modeled by static inhalation of carbon monoxide. The mice were randomized into three groups:DEACMP,control, and NBP.

  • Monoclonal antibodies like recaticimab target PCSK9 to effectively lower LDL cholesterol levels, offering an alternative to more frequent treatments due to its long-acting nature.
  • The study evaluated recaticimab's effectiveness and safety in patients with nonfamilial hypercholesterolemia and low-to-moderate atherosclerotic cardiovascular disease risk, comparing various dosing strategies.
  • Results showed significant LDL-C reductions (up to 52.8%) compared to placebo, with safety profiles similar to that of placebo, suggesting recaticimab may provide a flexible treatment option for patients.

Article Synopsis
  • There is a pressing need for new treatments for serious infections caused by Gram-negative bacteria, which the REVISIT study examined by evaluating aztreonam-avibactam against standard treatments in complicated infections.
  • The study involved a multinational, open-label design with adults who had complicated intra-abdominal infections or hospital-acquired pneumonia, comparing aztreonam-avibactam with meropenem or colistin over 5-14 days.
  • Results showed that out of 422 enrolled patients, 64% had identified Gram-negative pathogens, and the study aimed to determine clinical cure rates and safety without formal hypothesis testing.

Leukemia is a prevalent and widespread blood disease, and its early diagnosis is crucial for effective patient treatment. Diagnosing leukemia types heavily relies on pathologists' morphological examination of blood cell images. However, this process is tedious and time-consuming, and the diagnostic results are subjective, leading to potential misdiagnosis and underdiagnosis.

Background: Left ventricular hypertrophy (LVH) is most common when driven by hypertension, and it is a strong independent risk factor for adverse cardiovascular events and death. Some animal models support a role for gut microbiota and metabolites in the development of LVH, but cohort studies confirming these findings in populations are lacking.

Methods And Results: We investigated the alterations of gut microbiota and metabolites in 30 patients with hypertension, 30 patients with hypertensive LVH, and 30 matched controls on the basis of 16S rDNA and metabolomic analyses.

Background: The neurovascular unit (NVU) and neurovascular trophic coupling (NVTC) play a key regulatory role in brain injury caused by ischemic stroke. Salvianolic acids (SAL) and Panax notoginseng saponins (PNS) are widely used in China to manage ischemic stroke. Neuroprotective effects of SAL and PNS, either taken alone or in combination, were examined in this research.

Epigenetic regulation plays a role in Parkinson's disease (PD), and ten-eleven translocation methylcytosine dioxygenase 1 (TET1) catalyzes the first step in DNA demethylation by converting 5-methylcytosine to 5-hydroxymethylcytosine. We investigated whether TET1 binds to the promoter of the transient receptor potential cation channel subfamily V member 1 (TRPV1) and regulates its expression, thereby controlling oxidative stress in PD. TRPV1 was identified as an oxidative stress-associated gene in the GSE20186 dataset including substantia nigra from 14 patients with PD and 14 healthy controls and the Genecards database.

Function of NLRP3 inflammasome activation in multiple myeloma.

Hematology

December 2024

Department of Hematology, Baotou Central Hospital, Baotou, People's Republic of China.

Objective: The drug resistance of multiple myeloma (MM) cells is one of the main causes of relapse, refractory and progression of MM.

Methods: First, Western blot analysis was used to detect the expression levels of NLRP3, ASC, pro-IL-1β and cleaved IL-1β, and RT-qPCR was used to detect the mRNA expression levels of them. The expression levels of IL-1β and IL-18 in the supernatant were detected by ELISA, and the expression levels of these factors in the activated group and the control group were compared to verify the activation of BMMCs and KM3.
