Management of melanoma has changed significantly with the discovery of targeted therapies and immune checkpoint inhibitors (ICI). Our aim in the study is to determine which treatment alternatives, specifically dabrafenib plus trametinib and ICIs, are effective in adjuvant therapy and which treatment is effective as first-line metastatic therapy. This retrospective, multicenter study included 120 patients diagnosed with stage IIIB-IIID melanoma receiving both adjuvant and first-line metastatic treatment between 2007 and 2023.

Article Synopsis
  • Midostaurin is the first targeted treatment for AML FLT3 and reduces mortality by 23% compared to standard chemotherapy, but it has side effects, especially gastrointestinal issues.
  • A case was reported where a patient experienced acute pancreatitis symptoms after starting midostaurin, including abdominal pain and elevated enzyme levels.
  • The patient was treated with IV hydration and anti-symptomatic care, recovering after four days, but drug-induced acute pancreatitis remains a rare and complicated diagnosis without established links to midostaurin despite various potential mechanisms.

Article Synopsis
  • The study investigates the safety of omitting axillary lymph node dissection (ALND) in patients with residual axillary disease after neoadjuvant chemotherapy (NAC), focusing on those treated with sentinel lymph node biopsy (SLNB) or targeted axillary dissection (TAD).
  • An analysis of two multicenter cohorts (MF18-02 and MF18-03) included 501 patients who received regional nodal irradiation and achieved a complete clinical response to NAC.
  • Results showed low recurrence rates (0.4% axillary, 0.8% locoregional) and no significant differences in disease-free or disease-specific survival between SLNB and TAD, suggesting ALND omission is safe if patients receive

Optical imaging and gene transfection potential of linear polyethylenimine-coated AgS near-infrared quantum dots.

Turk J Biol

August 2024

Department of Chemistry, Koç University, İstanbul, Turkiye. Department of Pharmaceutical Biotechnology, Hamidiye Faculty of Pharmacy, University of Health Science, İstanbul, Turkiye. Experimental Medicine Practice and Research Center (EMPRC), Validebağ Research Center, University of Health Science, İstanbul, Turkiye.

Background/aim: The application of biocompatible heavy metal-free and cationic AgS NIR quantum dots (QDs), which have intense luminosity in the 700-900 nm medical range, as a nonviral gene delivery system paves the way to overcome autofluorescence and easily track the delivery of genes in real time.

Materials And Methods: The newly developed small and colloidally stable 2-mercaptopropionic acid (MPA)-capped AgS aqueous quantum dots electrostatically complexed with linear polyethyleneimine (AgS@2MPA/LPEI) were investigated for the first time both as a strong fluorescent probe and as a vector for nonviral gene delivery for the highest tracking of the system and delivery of genes into the nuclei of different cancer cells. The synthesized AgS@2MPA/LPEI quantum dots demonstrated strong optical imaging properties and were used to deliver a green fluorescent protein (GFP) plasmid as a standard gene.

The COVID-19 pandemic has profoundly affected healthcare systems, particularly post-stroke rehabilitation centers. The elevated severity of strokes and delayed hospital admissions caused numerous hurdles to rehabilitation administration during the peri-pandemic period. This study surveyed healthcare professionals' knowledge and perceptions of post-stroke rehabilitation during this period.

Background: Headache (HA) is the most common symptom impacting the quality of life of patients with Chiari I malformation. While HAs typically present in the occipital or suboccipital regions, and induced by Valsalva maneuver, complex migraine-like HAs referring to frontal or orbital regions also occur. Our study explores the therapeutic potential of C1 nerve root decompression in addition to increasing intracranial compliance in patients with Chiari I malformation.

Article Synopsis
  • The study investigated the effectiveness of ultrasound and CT scans for diagnosing salivary gland stones in 188 patients, with a focus on submandibular and parotid glands.
  • It found that CT scans identified some stones missed by ultrasound, and certain techniques in ultrasound can improve accuracy.
  • The research suggests avoiding CT for patients with dental work due to radiation concerns and recommends targeted ultrasound as a safer alternative.

Article Synopsis
  • - The study assessed trauma CT interpretation proficiency among emergency medicine residents in Turkey, finding a median accuracy rate of 64.9% with factors influencing interpretation performance analyzed.
  • - Four main factors linked to interpretation failure were identified: shorter interpretation times, lower self-confidence, over-reliance on radiology reports, and lack of departmental training programs.
  • - The findings suggest the necessity for improved educational strategies in radiology for emergency medicine residents to enhance their skills in trauma CT interpretation and ultimately improve patient care outcomes.

  • The study investigated the thickness of the Achilles tendon (ATT) and asprosin levels in women with polycystic ovary syndrome (PCOS) to explore links to cardio-metabolic diseases.
  • It included 45 PCOS patients and 30 healthy controls, measuring hormone levels and ATT using ultrasound to assess differences between the groups.
  • Results showed higher ATT and asprosin levels in PCOS patients, indicating a strong positive relationship, suggesting ATT measurement could be a simple tool for monitoring cardiometabolic health in these patients.

This bioequivalence research aims to evaluate the relative bioavailability and pharmacokinetic characteristics of ethinyl estradiol and drospirenone in the test preparation in comparison to the reference preparation during fasting conditions. A liquid chromatography method with tandem mass spectrometry was used to determine the concentrations of drospirenone and ethinyl estradiol in plasma. The pharmacokinetic parameters that were analyzed were the maximum plasma concentration (C), time to achieve C (t), elimination half life, and area under the concentration time curve of plasma (AUC, AUC for ethinyl estradiol, and AUC for drospirenone).

Widespread horse-based mobility arose around 2200 BCE in Eurasia.

Nature

July 2024

Centre d'Anthropobiologie et de Génomique de Toulouse, CNRS UMR 5288, Université Paul Sabatier, Faculté de Médecine Purpan, Toulouse, France.

Article Synopsis
  • - Horses transformed human mobility, but the timeline of their domestication and integration as transport is debated, with new genetic data being used to clarify this history.
  • - Analysis of 475 ancient horse genomes indicates that modern domestic horses were shaped by human intervention around 2200 BCE, after a domestication bottleneck began around 2700 BCE, leading to a significant expansion across Eurasia.
  • - Evidence also suggests that there was early horse husbandry in central Asia at Botai around 3500 BCE, prior to the establishment of contemporary horse bloodlines, challenging the notion of large herds being linked to migrations around 3000 BCE.

Article Synopsis
  • This study compares two surgical methods, Hartmann's procedure (HP) and resection with primary anastomosis (RPA), for treating acute left-sided colonic emergencies among 1215 patients from 204 centers globally.
  • Results showed that while HP was the more common treatment (57.3%), RPA was favored for younger patients with fewer health issues and those needing surgery sooner.
  • The study concluded that although HP is still widely used, RPA might be the better option, emphasizing the importance of patient characteristics and surgeon experience in determining treatment choice.
