Mosaic Analysis with Double Markers (MADM) represents a mouse genetic approach coupling differential fluorescent labeling to genetic manipulations in dividing cells and their lineages. MADM uniquely enables the generation and visualization of individual control or homozygous mutant cells in a heterozygous genetic environment. Among its diverse applications, MADM has been used to dissect cell-autonomous gene functions important for cortical development and neural development in general.

Determination of antimicrobial resistance (AMR) in pneumococcal isolates is important for surveillance purposes and in a clinical context. Antimicrobial susceptibility testing (AST) of pneumococci is complicated by the need for exact minimal inhibitory concentrations (MICs) of beta-lactam antibiotics. Two next-generation sequencing (NGS) analysis tools have implemented the prediction of AMR in their analysis workflow, including the prediction of MICs: Pathogenwatch (https://pathogen.

Laboratory Genes and Disease, Department of Laboratory Medicine, Medical University of Vienna (MUW), Vienna, Austria.

Genetically engineered mouse models (GEMMs) are instrumental for modelling local and systemic features of complex diseases such as cancer. Non-invasive, longitudinal cell detection and monitoring in tumors, metastases and/or the micro-environment is paramount to achieve a better spatiotemporal understanding of cancer progression and to evaluate therapies in preclinical studies. Bioluminescent and fluorescent reporters marking tumor cells or their microenvironment are valuable for non-invasive cell detection and monitoring in vivo.

Climate change poses an unprecedented threat to forest ecosystems, necessitating innovative adaptation strategies. Traditional assisted migration approaches, while promising, face challenges related to environmental constraints, forestry practices, phytosanitary risks, economic barriers, and legal constraints. This has sparked debate within the scientific community, with some advocating for the broader implementation of assisted migration despite these limitations, while others emphasize the importance of local adaptation, which may not keep pace with the rapid rate of climate change.

Available evidence suggests that various medical/rehabilitation treatments evoke multiple effects on blood hemostasis. It was therefore the aim of our study to examine whether fascial manipulation, vibration exercise, motor imagery, or neuro-muscular electrical stimulation can activate the coagulation system, and, thereby, expose patients to thrombotic risk. Ten healthy young subject were enrolled in the study.

Purpose Of Review: Men face distinctive health-related challenges as a result of biological, behavioral, and sociocultural factors. In addition, the modern healthcare system does not offer men equal opportunities and options to ensure sex-specific access and delivery to health services. Men's health concerns are, indeed, often not addressed or even forgotten.

  • The study evaluated the impact of judo on neurodevelopment in young children aged 4-7 years, comparing kids who practiced judo with those who did not over a 6-month period.
  • Participants were assessed using specific developmental measurement tools, indicating that judo may enhance both cognitive and motor skills, as demonstrated by significant improvements in the judo group.
  • Results suggest that incorporating judo into early childhood education could support central nervous system maturation, although the screening tool used for parent evaluations was less effective.

January 2025

Medical Department of Hematology, Oncology and Tumor Immunology, Molekulares Krebsforschungszentrum - MKFZ, Campus Virchow Klinikum, Charité - Universitätsmedizin, Berlin, Germany.

Article Synopsis
  • Therapy-exposed surviving cancer cells can undergo significant changes in their epigenetics, making them more resilient and likely to cause aggressive relapses.
  • Ramponi and colleagues researched a specific type of cell behavior in lung cancer and melanoma, induced by mTOR/PI3K inhibitors, noting how these cells mimic some characteristics of senescent cells but lack an inflammatory response typical of those cells.
  • Their findings indicate potential weaknesses in these drug-tolerant cells and highlight challenges in studying these behaviors in laboratory settings, sparking discussions on the nature and treatment of persister cells in cancer.

Multifunctional ortho-quinones are required for the formation of thiol-catechol-connectivities (TCC) but can be delicate to handle. We present the electrochemical oxidation of the dipeptide DiDOPA, achieving up to 92% conversion efficiency of the catechols to ortho-quinones. Graphite and stainless steel could be employed as cost-efficient electrodes.

  • Amniotic epithelial stem cells (AEC) have regenerative potential, particularly through their conditioned medium (AEC-CM), which shows immunomodulatory and regenerative effects.
  • A novel hydrogel made from hyaluronic acid and polyethylene glycol was developed to encapsulate AEC-CM, with features like fast cross-linking and sustained growth factor release over 14 days.
  • Studies demonstrate that the encapsulated AEC-CM significantly enhances the suppression of peripheral blood mononuclear cells and promotes a shift towards the beneficial M2 macrophage phenotype, suggesting its viability as an immunomodulatory treatment for tissue regeneration.

January 2025

Center for Dental Medicine, Department of Operative Dentistry and Periodontology, Faculty of Medicine and Medical Center, University of Freiburg, Hugstetter Str. 55, 79106, Freiburg im Breisgau, Germany.

Objectives: The study aimed to assess the percent correct shade identification of four intraoral scanners (IOS) and a spectrophotometer, focusing on how reliably each device selects the correct tooth shade compared to a visual observer's selection. The research question addresses how much clinicians can trust the device-selected shade without visual verification.

Materials And Methods: Sixteen participants with natural, unrestored teeth were included.

In organisms ranging from vertebrates to plants, major components of centromeres are rapidly evolving repeat sequences, such as tandem repeats (TRs) and transposable elements (TEs), which harbour centromere-specific histone H3 (CENH3). Complete centromere structures recently determined in human and Arabidopsis suggest frequent integration and purging of retrotransposons within the TR regions of centromeres. Despite the high impact of 'centrophilic' retrotransposons on the paradox of rapid centromere evolution, the mechanisms involved in centromere targeting remain poorly understood in any organism.

Department of Laboratory Medicine, Medical University of Vienna, Vienna, Austria.

Article Synopsis
  • ApoB is the key protein found in LDL and other lipoproteins, playing a major role in their formation and link to atherosclerosis.
  • LDL contributes to plaque formation in arteries by entering the wall and triggering inflammatory responses through interaction with other molecules, leading to harmful processes like foam cell formation.
  • Research has identified potential interventions to combat atherosclerosis by lowering lipoprotein levels and addressing the inflammatory responses in the arterial wall.

State Key Laboratory of Rice Biology and Breeding; Ministry of Agriculture Key Lab of Molecular Biology of Crop Pathogens and Insects; Zhejiang Provincial Key Laboratory of Biology of Crop Pathogens and Insects; Institute of Biotechnology, Zhejiang University, Hangzhou 310058, China.

Crop production currently relies on the widespread use of agrochemicals to ensure food security. This practice is considered unsustainable, yet has no viable alternative at present. The plant microbiota can fulfil various functions for its host, some of which could be the basis for developing sustainable protection and fertilization strategies for plants without relying on chemicals.

In this brief contribution-happily afforded by the 20th anniversary of Physics of Life Reviews-we take the opportunity to reflect on our earlier published paper, which had introduced a theoretical framework, the VIMAP (Vienna Integrated Model of (Top-Down and Bottom-up processes in) Art Perception; (Pelowski et al., [1])) that has come to represent a major basis for organizing, anticipating, and empirically investigating the nuanced, multivariate visual art experience. We look back at the original model and its hypotheses, especially as these regard distinct "outcomes," which we had argued may provide a superstructure of supraordinate, shared varieties of art experience detected across individual meetings of viewer, context, and artworks.
