Liquid biopsy provides a minimally invasive approach to characterise the molecular and phenotypic characteristics of a patient's individual tumour by detecting evidence of cancerous change in readily available body fluids, usually the blood. When applied at multiple points during the disease journey, it can be used to monitor a patient's response to treatment and to personalise clinical management based on changes in disease burden and molecular findings. Traditional liquid biopsy approaches such as quantitative PCR, have tended to look at only a few biomarkers, and are aimed at early detection of disease or disease relapse using predefined markers.

Since May 2024, New Caledonia has faced civil unrest, economic collapse, food insecurity, and social instability, severely disrupting environmental management in this globally significant biodiversity hotspot. The crisis has exacerbated threats to biodiversity from poaching, illegal fishing, deforestation, urban fires, waste pollution, and pet abandonment, while conservation efforts have ground to a halt. Understanding Autism as a Condition in Mental Health Clinical Practice: Clinical Perspectives from a Youth Early Psychosis Service.

Autism is a rapidly growing phenomenon, with rates of diagnosed autism in the community rising every decade. Autism and traits of autism are also regularly part of presentation at youth mental health services, including early psychosis services. In early psychosis services young people's symptoms tend to be formulated through a psychosis lens, rather than a neurodevelopmental lens which can lead to unnecessary medicalised treatment, and treatment plans that do not consider the possible impact of neurodiversity.

Background: Using the lived experience as their expertise, peer support specialists (PSS) focus on the empowerment of individuals with mental health conditions. Despite its value, peer support services in Singapore are relatively uncommon. Despite extensive literature, it is unclear whether smokeless tobacco consumption during pregnancy has an adverse effect on birth weight.

O-Fucosylation plays crucial roles in various essential biological events. Alongside the well-established O-fucosylation of epidermal growth factor-like repeats by protein O-fucosyltransferase 1 (POFUT1) and thrombospondin type 1 repeats by POFUT2, we recently identified a type of O-fucosylation on the elastin microfibril interface (EMI) domain of Multimerin-1 (MMRN1). Here, using AlphaFold2 screens, co-immunoprecipitation, enzymatic assays combined with mass spectrometric analysis and CRISPR-Cas9 knockouts, we demonstrate that FUT10 and FUT11, originally annotated in UniProt as α1,3-fucosyltransferases, are actually POFUTs responsible for modifying EMI domains; thus, we renamed them as POFUT3 and POFUT4, respectively.

Purpose Of Review: Lowering low-density lipoprotein (LDL)-cholesterol reduces cardiovascular risk. International lipid management guidelines recommend LDL-cholesterol goals or thresholds for initiating lipid-lowering therapy. However, contemporary real-world studies have shown that many high- and very high-risk patients are not attaining LDL-cholesterol goals and are not receiving intensive lipid-lowering therapies.

Background: Remote monitoring using electronic patient-reported outcomes (ePROs) may help identify immune-related adverse events (irAEs) and direct self-management. There is no consensus regarding thresholds to alert providers about potentially severe irAEs or when to instigate evidence-based self-management. The click beetles (Elateridae) represent the major and well-known group of the polyphagan superfamily Elateroidea. Despite a relatively rich fossil record of Mesozoic Elateridae, only a few species are described from the Upper Cretaceous Burmese amber. Although Elateridae spend most of their lives as larvae, our knowledge on immature stages of this family is limited, which is especially valid for the fossils.

Low-dimensional controllability of brain networks.

Identifying the driver nodes of a network has crucial implications in biological systems from unveiling causal interactions to informing effective intervention strategies. Despite recent advances in network control theory, results remain inaccurate as the number of drivers becomes too small compared to the network size, thus limiting the concrete usability in many real-life applications. To overcome this issue, we introduced a framework that integrates principles from spectral graph theory and output controllability to project the network state into a smaller topological space formed by the Laplacian network structure.

Identification of Environmental Determinants Involved in the Distribution of Burkholderia pseudomallei in Southeast Asia using MaxEnt software.

Burkholderia pseudomallei (Bp), causing melioidosis, is becoming a major global public health concern. It is highly endemic in Southeast Asia (SEA) and Northern Australia and is persisting beyond the established areas of endemicity. This study aimed to determine the environmental variables that would predict the most suitable ecological niche for this pathogenic bacterium in SEA by maximum entropy (MaxEnt) modeling.

Intracerebroventricular B7-H3-targeting CAR T cells for diffuse intrinsic pontine glioma: a phase 1 trial.

Diffuse intrinsic pontine glioma (DIPG) is a fatal central nervous system (CNS) tumor that confers a median survival of 11 months. As B7-H3 is expressed on pediatric CNS tumors, we conducted BrainChild-03, a single-center, dose-escalation phase 1 clinical trial of repetitive intracerebroventricular (ICV) dosing of B7-H3-targeting chimeric antigen receptor T cells (B7-H3 CAR T cells) for children with recurrent or refractory CNS tumors and DIPG. Here we report results from Arm C, restricted to patients with DIPG.

Neoadjuvant immunotherapies have shown antitumor activity in melanoma. Substudy 02C of the global, rolling-arm, phase 1/2, adaptive-design KEYMAKER-U02 trial is evaluating neoadjuvant pembrolizumab (anti-PD-1) alone or in combination, followed by adjuvant pembrolizumab, for stage IIIB-D melanoma. Here we report results from the first three arms: pembrolizumab plus vibostolimab (anti-TIGIT), pembrolizumab plus gebasaxturev (coxsackievirus A21) and pembrolizumab monotherapy.

Inferring effects of mutations on SARS-CoV-2 transmission from genomic surveillance data.

New and more transmissible variants of SARS-CoV-2 have arisen multiple times over the course of the pandemic. Rapidly identifying mutations that affect transmission could improve our understanding of viral biology and highlight new variants that warrant further study. Here we develop a generic, analytical epidemiological model to infer the transmission effects of mutations from genomic surveillance data.

Increasing photosynthetic benefit with decreasing irrigation frequency in an Australian temperate pasture exposed to elevated carbon dioxide.

Elevated atmospheric CO2 (e[CO2]) often enhances plant photosynthesis and improves water status. However, the effects of e[CO2] vary significantly and are believed to be influenced by water availability. With the future warmer climate expected to increase the frequency and severity of extreme rainfall, the response of plants to e[CO2] under changing precipitation patterns remains uncertain.

Activation of the kynurenine pathway identified in individuals with covert hepatic encephalopathy.

Background: HE is a neuropsychiatric complication of liver disease characterized by systemic elevation in ammonia and proinflammatory cytokines. These neurotoxins cross the blood-brain barrier and cause neuroinflammation, which can activate the kynurenine pathway (KP). This results in dysregulated production of neuroactive KP metabolites, such as quinolinic acid, which is known to cause astrocyte and neuronal death.
