Lamellarity of ultrasound assisted formations of dipalmitoyl-lecithin vesicles.

Ultrason Sonochem

December 2024

Research Centre for Natural Sciences, Institute of Materials and Environmental Chemistry, Magyar tudósok körútja 2, 1117 Budapest, Hungary.

Formation of unilamellae of fully hydrated dipalmitoylphosphatidylcholine (DPPC) was induced by a horn sonicator from multilamellar vesicles and followed by time-resolved synchrotron small angle X-ray scattering and direct visual morphological investigations by the means of transmission electron-microscopy combined with freeze-fracture. Without incubation the ultrasonication causes continuous increasing in temperature and transformation from the gel to rippled gel structures, then reaching the main transition, the formfactor of unilamellar structure appeared. The ultrasonication resulted in different layer formations at the characteristic temperatures of the gel (20 °C), rippled gel (38 °C), and liquid crystalline (45 °C) phases of the system.

Peritoneal dialysis (PD) is an increasingly needed, life-maintaining kidney replacement therapy; efficient solute transport is critical for patient outcome. While the role of peritoneal perfusion on solute transport in PD has been described, the role of cellular barriers is uncertain, the mesothelium has been considered irrelevant. We calculated peritoneal blood microvascular endothelial (BESA) to mesothelial surface area (MSA) ratio in human peritonea in health, chronic kidney disease, and on PD, and performed molecular transport related gene profiling and single molecule localization microscopy in two mesothelial (MC) and two endothelial cell lines (EC).

NLRP3 inflammasome mediates astroglial dysregulation of innate and adaptive immune responses in schizophrenia.

Brain Behav Immun

November 2024

Department of Medical Genetics, Oslo University Hospital, Oslo, Norway; K.G. Jebsen Centre for Neurodevelopmental Disorders, University of Oslo, Oslo, Norway; Department of Clinical Science, University of Bergen, Bergen, Norway.

Mounting evidence indicates the involvement of neuroinflammation in the development of schizophrenia (SCZ), but the potential role of astroglia in this phenomenon remains poorly understood. We assessed the molecular and functional consequences of inflammasome activation using induced pluripotent stem cell (iPSC)-derived astrocytes generated from SCZ patients and healthy controls (CTRL). Screening protein levels in astrocytes at baseline identified lower expression of the NLRP3-ASC complex in SCZ, but increased Caspase-1 activity upon specific NLRP3 stimulation compared to CTRL.

One of the major risks associated with the concomitant use of herbal products and therapeutic drugs is herb-drug interactions (HDIs). The most common mechanism leading to HDIs is the inhibition and/or induction of transport proteins and drug-metabolizing enzymes by herbal ingredients, causing changes in the pharmacokinetic disposition of the victim drug. The present study aimed to determine the potential interactions of (UT) (cat's claw), a popular herb due to its supposed health benefits.

For mitogen-activated protein kinases (MAPKs) a shallow surface-distinct from the substrate binding pocket-called the D(ocking)-groove governs partner protein binding. Screening of broad range of Michael acceptor compounds identified a double-activated, sterically crowded cyclohexenone moiety as a promising scaffold. We show that compounds bearing this structurally complex chiral warhead are able to target the conserved MAPK D-groove cysteine via reversible covalent modification and interfere with the protein-protein interactions of MAPKs.
