Background: Currently, acute ischemic stroke is still a leading cause of mortality and morbidity. Approximately 2 years ago, mechanical thrombectomy was proven beneficial as a revolutionary new therapy for stroke in the MR-CLEAN trial (A Multicenter Randomized Clinical Trial of Endovascular Treatment for Acute Ischemic Stroke in the Netherlands). However, the mechanisms by which the thrombectomy device, or stent-retriever, interacts with the thrombus are largely unknown.

Whole-Exome Sequencing in Age-Related Macular Degeneration Identifies Rare Variants in COL8A1, a Component of Bruch's Membrane.

Ophthalmology

September 2018

Department of Ophthalmology, Donders Institute for Brain, Cognition and Behaviour, Radboud University Medical Center, Nijmegen, The Netherlands; Department of Human Genetics, Donders Institute for Brain, Cognition and Behaviour, Radboud University Medical Center, Nijmegen, The Netherlands. Electronic address:

Purpose: Genome-wide association studies and targeted sequencing studies of candidate genes have identified common and rare variants that are associated with age-related macular degeneration (AMD). Whole-exome sequencing (WES) studies allow a more comprehensive analysis of rare coding variants across all genes of the genome and will contribute to a better understanding of the underlying disease mechanisms. To date, the number of WES studies in AMD case-control cohorts remains scarce and sample sizes are limited.

Genetic correlation of antisocial behaviour with alcohol, nicotine, and cannabis use.

Drug Alcohol Depend

June 2018

Behavioural Science Institute, Radboud University, Montessorilaan 3, 6525 HR Nijmegen, The Netherlands; Department of Biological Psychology/Netherlands Twin Register, VU University, van der Boechorststraat 1, 1081 BT Amsterdam, The Netherlands; Department of Psychiatry, Amsterdam Medical Center, University of Amsterdam, Meibergdreef 5, 1105 AZ Amsterdam, The Netherlands. Electronic address:

Background: There is high comorbidity between antisocial behaviour (ASB) and substance use, and twin studies have shown that part of the covariation is due to overlapping genetic influences. Here we used measured genetic effects to estimate the genetic correlations of ASB with nicotine, alcohol, and cannabis use.

Methods: We meta-analysed data from two genome-wide association studies for ASB and used existing summary statistics from the largest genome-wide association studies into substance use (ever smoking, cigarettes smoked per day, weekly alcohol consumption, and lifetime cannabis use).

Reproductive intentions and use of reproductive health care among female survivors of childhood cancer.

Hum Reprod

June 2018

Department of Paediatrics, Division of Oncology-Haematology, VU University Medical Center, PO Box 7057, 1000 MB Amsterdam, The Netherlands.

Study Question: Do female childhood cancer survivors (CCSs) express a decreased desire to have children and do they use reproductive health care more often compared to women without a history of cancer?

Summary Answer: Overall, no difference was found in the desire to have children between CCSs and controls, whereas CCSs consult a fertility specialist more often, at a younger age, and sooner after their first attempt at conceiving.

What Is Known Already: Female CCSs may face a shorter than anticipated reproductive window as a result of their cancer treatment. Little is known about their desire to have children and use of reproductive health care, especially in relation to their former cancer treatment.

Article Synopsis
  • The study investigates GS-458967 (GS967), a selective inhibitor of late sodium current, for its ability to prevent Torsades de Pointes (TdP) arrhythmias in dogs with chronic atrioventricular block.
  • Experimental results show that GS967 effectively shortens repolarization in cardiac cells and completely eliminates TdP in most cases, while some early afterdepolarizations remain unaffected.
  • The findings suggest GS967 works by reducing spatial dispersion of repolarization, thereby interrupting the progression of arrhythmias rather than stopping their initial triggers.

Article Synopsis
  • Vanishing white matter (VWM) is a severe genetic disorder in children, leading to neurological decline and often resulting in death within a few years after symptoms appear.
  • The disease stems from mutations in the eIF2B gene, which is crucial for regulating protein synthesis in cells.
  • A study using a mouse model of VWM investigated whether the astrocytes (a type of brain cell) in these mice showed an exaggerated response to stress, but results indicated that these cells did not exhibit a hyperactive stress response as initially hypothesized.

Article Synopsis
  • MRI is recommended for early diagnosis of rheumatoid arthritis (RA) to detect joint damage, but symptom-free individuals, especially the elderly, can also show similar erosions.
  • A study compared MRI results of 589 early arthritis patients (238 with RA) and 193 symptom-free controls, focusing on erosion scores and inflammation in the joints.
  • Results indicated that while RA patients had significantly higher erosion scores, the specific erosion characteristics were infrequently present, suggesting the need for caution in distinguishing RA from other conditions based solely on MRI findings.
