Cushing syndrome.

Nat Rev Dis Primers

January 2025

Endocrine Division, Department of Medicine, Centre hospitalier de l'Université de Montréal (CHUM), Montreal, Québec, Canada.

Cushing syndrome (CS) is a constellation of signs and symptoms caused by excessive exposure to exogenous or endogenous glucocorticoid hormones. Endogenous CS is caused by increased cortisol production by one or both adrenal glands (adrenal CS) or by elevated adrenocorticotropic hormone (ACTH) secretion from a pituitary tumour (Cushing disease (CD)) or non-pituitary tumour (ectopic ACTH secretion), which stimulates excessive cortisol production. CS is associated with severe multisystem morbidity, including impaired cardiovascular and metabolic function, infections and neuropsychiatric disorders, which notably reduce quality of life.

Sex-specific DNA methylation marks associated with sex-biased risk of recurrence in unprovoked venous thromboembolism.

J Thromb Haemost

January 2025

Dalla Lana School of Public Health, University of Toronto, Toronto, Canada; Office of the Vice-Principal of Research and Innovation, University of Toronto Mississauga, Mississauga, Canada. Electronic address:

Background: Whether to stop oral anticoagulants after a first unprovoked venous thromboembolism (VTE) is challenging, partially due to an intriguingly higher risk of VTE recurrence (rVTE) in men after therapy discontinuation. DNA methylation (DNAm) differences between men and women might underly this sex-biased rVTE risk difference.

Aim: To investigate sex-specific associations between DNAm at cytosine-phosphate-guanine (CpG) sites and rVTE.

Phenotypic Heterogeneity of ADTKD-MUC1 Diagnosed Using VNtyper, a Novel Genetic Technique.

Am J Kidney Dis

January 2025

Hereditary Kidney Diseases Laboratory, Inserm UMR 1163, Imagine Institute, Paris Cité University, Paris, France; Department of Genomic Medicine for Rare Diseases, Necker-Enfants Malades Hospital, Assistance publique, Hôpitaux de Paris (AP-HP), Paris, France. Electronic address:

Rationale & Objective: Molecular diagnosis of autosomal dominant tubulointerstitial kidney disease (ADTKD) due to variants in the MUC1 gene has long been challenging since variants lie in a large Variable Number of Tandem Repeat (VNTR) region, making identification impossible using standard short read techniques. Previously, we addressed this diagnostic limitation by developing a computational pipeline, named VNtyper, for easier reliable detection of MUC1 VNTR pathogenic variants from short read sequences. This led to unexpected diagnoses of ADTKD-MUC1 among patients with kidney disease referred for genetic testing, which we report here.

Drought stress during the plant's growing season is a serious constraint to plant establishment in arid and semiarid Mediterranean ecosystems. Plant growth promoting rhizobacteria (PGPR) as environmentally friendly and innovative management approach can be used to produce seedlings better adapted to these environments. We tested native PGPR strains isolated from drought-tolerant tree and shrub species originating from two climatically contrasting regions: hot-dry (Dehloran) and milder Mediterranean climate (Ilam).

Modeling impairment of ionic regulation with extended Adaptive Exponential integrate-and-fire models.

J Comput Neurosci

January 2025

Paris-Saclay University, Centre National de la Recherche Scientifique (CNRS), Institute of Neuroscience (NeuroPSI), 91198, Gif sur Yvette, France.

To model the dynamics of neuron membrane excitability many models can be considered, from the most biophysically detailed to the highest level of phenomenological description. Recent works at the single neuron level have shown the importance of taking into account the evolution of slow variables such as ionic concentration. A reduction of such a model to models of the integrate-and-fire family is interesting to then go to large network models.

January 2025

Translational Neuroimaging Group, Center for Image Sciences, University Medical Center Utrecht, Utrecht University, Utrecht, The Netherlands.

Blood flow velocity in the cerebral perforating arteries can be quantified in a two-dimensional plane with phase contrast magnetic imaging (2D PC-MRI). The velocity pulsatility index (PI) can inform on the stiffness of these perforating arteries, which is related to several cerebrovascular diseases. Currently, there is no open-source analysis tool for 2D PC-MRI data from these small vessels, impeding the usage of these measurements.

CprA is a short-chain dehydrogenase/reductase (SDR) that contributes to resistance against colistin and antimicrobial peptides. The cprA gene is conserved across Pseudomonas aeruginosa clades and its expression is directly regulated by the two-component system PmrAB. We have shown that cprA expression leads to the production of outer membrane vesicles (OMVs) that block autophagic flux and have a greater capacity to activate the non-canonical inflammasome pathway.

We investigated the reactivity of a -dichlorovinyl-carbazole precursor in the on-surface synthesis approach. Our findings reveal that, on the Au(111) surface, the thermally-induced dehalogenation reaction led to the formation of cumulene dimers. Contrastingly, the more reactive Cu(111) surface promoted the formation of a polyheterocyclic compound exhibiting extended aromaticity.

Synthesis and Chiroptical Activity of π-Expanded Electron-rich Heterohelicenes Based on the 1,4-Dihydropyrrolo[3,2-b]pyrrole core.

Chemistry

January 2025

Institute of Organic Chemistry PAS: Instytut Chemii Organicznej Polskiej Akademii Nauk, Institute of Organic Chemistry, Kasprzaka 44/52, 01-224, Warsaw, POLAND.

Herein, we report the synthesis and chiroptical characteristics of the first (double) helicenes possesing the 1,4-dihydropyrrolo[3,2-b]pyrrole (DHPP) moiety as their central core. We have developed a three-step synthesis with 6π-electrocyclization accompanied with HBr elimination as its key step. We found that, whereas for smaller periphereal arms double 6π-electrocyclization occurs smoothly forming a double helicene, in the case of longer policyclic aromatic hydrocarbons the reaction becomes less efficient and mono-helicenes are the only products.

A novel coordination motif comprising [4]helicene fused with pyrazino-phenanthroline (H4PP) has been synthesized and reacted with ReCl(CO)5 to yield its rhenium(I) complex (Re-H4PP). Absorption and emission spectroscopic analysis conducted in dichloromethane and 2-methyltetrahydrofuran reveals that combining pyrazino-phenanthroline with helicene visibly affects the photophysical attributes of both the resulting ligand and its Re(I) complex as compared to their non-helicene analogues, and even more importantly leads to relatively high photoluminescence quantum yield values, especially in the case of H4PP (29%). Chiroptical studies through electronic circular dichroism and circularly polarized luminescence performed on enantiomerically enriched samples of Re-H4PP show the chiral nature of low-energy excited states affording notable glum values that amplify at cryogenic temperatures.
