Background: One of the key steps in arthroscopic Anterior Cruciate Ligament Reconstruction (ACLR) is getting the femoral tunnel at the right position to attach the graft. While the correct position has been described as a low and posterior position behind the bifurcate ridge on the medial surface of lateral femoral condyle, to reproducibly achieve it more than one technique is being used by surgeons. There are no randomized studies in literature which have evaluated the efficacy of these in a surgeon's hand.

The role of macrophage and adipocyte mitochondrial dysfunction in the pathogenesis of obesity.

Front Immunol

November 2024

Department of Laboratory Medicine, West China Second University Hospital, Sichuan University, Chengdu, Sichuan, China.

Obesity has emerged as a prominent global public health concern, leading to the development of numerous metabolic disorders such as cardiovascular diseases, type-2 diabetes mellitus (T2DM), sleep apnea and several system diseases. It is widely recognized that obesity is characterized by a state of inflammation, with immune cells-particularly macrophages-playing a significant role in its pathogenesis through the production of inflammatory cytokines and activation of corresponding pathways. In addition to their immune functions, macrophages have also been implicated in lipogenesis.

Specific ECM degradation potentiates the antitumor activity of CAR-T cells in solid tumors.

Cell Mol Immunol

December 2024

State Key Laboratory of Holistic Integrative Management of Gastrointestinal Cancers, Department of Biochemistry and Molecular Biology, Fourth Military Medical University, Xi'an, Shaanxi, China.

Although major progress has been made in the use of chimeric antigen receptor (CAR)-T-cell therapy for hematological malignancies, this method is ineffective against solid tumors largely because of the limited infiltration, activation and proliferation of CAR-T cells. To overcome this issue, we engineered CAR-T cells with synthetic Notch (synNotch) receptors, which induce local tumor-specific secretion of extracellular matrix (ECM)-degrading enzymes at the tumor site. SynNotch CAR-T cells achieve precise ECM recognition and robustly kill targeted tumors, with synNotch-induced enzyme production enabling the degradation of components of the tumor ECM.
