Over the past few years, there has been an increasing interest in making oligonucleotides specific for ammonia-oxidizing bacteria (AOB), in order to detect and monitor these slow growing bacteria in environmental samples, in enrichment cultures and in wastewater treatment plants. Based on 16S rDNA sequences, a broad selection of oligonucleotides have been designed, either encompassing all known AOB in the beta-subgroup of the Proteobacteria (beta AOB), or subclasses within beta AOB. Thirty different oligonucleotides have so far been published, with varying specificity.

Two isoforms of peroxisome proliferator-activated receptor gamma (PPAR gamma) cDNAs, gamma 1 and gamma 2, have been isolated and characterised in swine. The relative expression of the two transcripts was studied by northern blot analysis using total RNA isolated from several porcine tissues taken at three different ages (day 1, after 5 weeks and at 100 kg weight). Hybridisation were carried out with two different probes, one binding to both PPAR gamma transcripts and the other being PPAR gamma 2 specific.

Four class IIa bacteriocins (pediocin PA-1, enterocin A, sakacin P, and curvacin A) were purified to homogeneity and tested for activity toward a variety of indicator strains. Pediocin PA-1 and enterocin A inhibited more strains and had generally lower MICs than sakacin P and curvacin A. The antagonistic activity of pediocin-PA1 and enterocin A was much more sensitive to reduction of disulfide bonds than the antagonistic activity of sakacin P and curvacin A, suggesting that an extra disulfide bond that is present in the former two may contribute to their high levels of activity.

Rosette-formed, circular thalli of Degelia plumbea were studied in the laboratory. Regardless of thallus size, the optimal quantum yield of photosystem II (F /F ) remained at a high, constant level during a drying cycle starting with fully hydrated thalli until the thallus water content reached about 200%. Net photosynthesis reached a maximum level at this hydration level.

Proteins from Clostridium pasteurianum were separated by two-dimensional gel electrophoresis. Amino-terminal sequence determination and sequence analysis allowed the identification of 20 proteins, while 11 protein sequences remained unidentified and one protein appeared to have a blocked amino terminus.

We studied the kinetics of lysine, methionine, and threonine in six high-yielding dairy cows at peak lactation (stage 1) and 6 mo later (stage 2). The cows were fitted with cannulas in the rumen and duodenum and were automatically fed every 4th h. The three amino acids (AA) were administered intraruminally in mixtures at dosages of 100, 200, 300, and 400 mmol of each, together with polyethylene glycol (PEG) as a liquid marker.

A wide range of complex systems appear to have switch-like interactions, i.e. below (or above) a certain threshold x has no or little influence on y, while above (or below) this threshold the effect of x on y saturates rapidly to a constant level.

Production of the bacteriocins enterocin A and enterocin B in Enterococcus faecium CTC492 was dependent on the presence of an extracellular peptide produced by the strain itself. This induction factor (EntF) was purified, and amino acid sequencing combined with DNA sequencing of the corresponding gene identified it as a peptide of 25 amino acids. The gene encodes a prepeptide of 41 amino acids, including a 16-amino-acid leader peptide of the double-glycine type.

In two experiments, no consistent differences in weight gain or food intake appeared between broiler chickens given diets based on whole or rolled barley, but food: gain ratio and gizzard weight as a percentage of live weight were reduced (P < 0.05) when barley was rolled.

Production of the bacteriocin sakacin P by Lactobacillus sake LTH673 is dependent on a secreted 19-residue peptide pheromone (IP-673). The gene encoding IP-673 (sppIP) was identified and sequenced. SppIP was shown to be co-transcribed with genes encoding a histidine kinase (sppK) and a response regulator (sppR) typical for signal transduction in bacteria.

ADP-glucose pyrophosphorylase from photosynthetic tissue is allosterically regulated by 3-phosphoglycerate and inorganic phosphate. In contrast, data from our laboratory indicated that the major AGPase from barley seeds is insensitive to these regulators. Verification of this conclusion has, however, been hindered by the proteolytic degradation of the enzyme from seeds.

Heat processing endpoint temperature affects several quality parameters of meat. The present paper focuses on using near-infrared and visual light reflectance spectroscopy at 400-2500 nm to determine the endpoint temperature in previously heated ground beef. Glass fibre filters with beef juice centrifuged from m.

In this report we describe the isolation and characterisation of the cDNAs encoding two isoforms of peroxisome proliferator-activated receptor gamma (PPAR gamma), gamma 1 and gamma 2, in cattle. The cDNA sequences show strong conservation with the corresponding sequences reported in other species. The distribution of PPAR gamma mRNAs in various bovine tissues was investigated using Northern blot analysis.

To map the incidence of natural competence in the genus Streptococcus, we used PCR to screen a number of streptococcal strains for the presence of the recently identified competence regulation operon, containing the comC, -D, and -E genes. This approach established that the operon is present in strains belonging to the S. mitis and S.

Russian-Norwegian expeditions to the Kara Sea and to dumping sites in the fjords of Novaya Zemlya have taken place annually since 1992. In the fjords, dumped objects were localised with sonar and ROV equipped with underwater camera. Enhanced levels of 137Cs, 60Co, 90Sr and 239,240Pu in sediments close to dumped containers in the Abrosimov and Stepovogo fjords demonstrated that leaching from dumped material has taken place.

A full length cDNA clone encoding the 447 amino acid long barley (Hordeum vulgare cv. Bomi) endosperm elongation factor 1 alpha (eF-1 alpha) was isolated by a differential screening procedure. RFLP mapping of eF-1 alpha showed that the barley genome contains a small eF-1 alpha gene family of 4 copies, with 1 copy of the gene being located on each of chromosomes 2, 4, 6, and 7.

Polymorphic sites within the bovine FcgammaRI (FCGR1), FcgammaRII (FCGR2), and FcgammaRIII (FCGR3) genes were used for proximal mapping of these genes to bovine Chromosome (Chr) 3 (BTA3) with paternal half-sib families from Norwegian Cattle. A fine-structure genetic map of the region was obtained by the analysis of 288 sperm cells from three bulls that were heterozygous for the loci included in the study. No recombinants were observed between FCGR2 and FCGR3 (242 sperm cells).

This report summarizes the results from an effort to optimize the double-agar plate assay for visualization of the plaques made by six temperate bacteriophages induced from industrial strains of Lactococcus lactis. Among the several parameters found to influence the plaque assay, the effect of incorporating glycine into the growth medium was most striking, resulting in extensive increase in the plaque size of all of the 13 phage-host pairs tested. Notable effects on the plaque size of other factors such as the procedure for sterilization of the agar medium, the volume and softness of the top and bottom layers, and the number and growth stage of the bacterial cells added to the lawn, were also observed.

The strain Enterococcus faecium T136 produces two bacteriocins, enterocin A, a member of the pediocin family of bacteriocins, and a new bacteriocin termed enterocin B. The N-terminal amino acid sequences of enterocins A and B were determined, and the gene encoding enterocin B was sequenced. The primary translation product was a 71 aa peptide containing a leader peptide of the double-glycine type which is cleaved off to give mature enterocin B of 53 aa.
