CCL3 in the bone marrow microenvironment causes bone loss and bone marrow adiposity in aged mice.

JCI Insight

January 2023

Shanghai Key Laboratory of Orthopedic Implants, Department of Orthopedic Surgery, Shanghai Ninth People's Hospital, Shanghai Jiao Tong University School of Medicine, Shanghai, China.

Article Synopsis
  • The study explores how aging affects bone marrow stromal cells (BMSCs), showing that older BMSCs tend to turn into fat cells (adipocytes) rather than bone cells (osteocytes).
  • Researchers found that CC chemokine ligand 3 (CCL3) accumulates in aged mice's blood, contributing to the negative changes in bone health, such as bone loss and increased fat in the bone marrow.
  • By blocking or deleting CCL3, the study revealed potential ways to counteract these age-related issues in bone health, suggesting new treatment targets for osteoporosis in the elderly.

Role of MMP-2 and CD147 in kidney fibrosis.

Open Life Sci

September 2022

Department of Pathology and Pathophysiology, Medical School, Southeast University, Dingjiaqiao 87, Gulou District, Nanjing 210009, Jiangsu Province, China.

Matrix metalloproteinase-2 (MMP-2) and cluster of differentiation 147 (CD147) both play important roles in the development of kidney fibrosis, and CD147 can induce the production and activation of MMP-2. In the early stage of kidney fibrosis, MMP-2 promotes extracellular matrix (ECM) production and accelerates the development of kidney fibrosis, while in the advanced stage, MMP-2 activity decreases, leading to reduced ECM degradation and making it difficult to alleviate kidney fibrosis. The reason for the decrease in MMP-2 activity in the advanced stage is still unclear.

The interaction between over-proliferating keratinocytes and excessive activated immune cells contributes to the development and progression of psoriasis. Photodynamic therapy (PDT)/photothermal therapy (PTT) is a promising method to treat skin diseases due to their synergistic effect and low side effect. Cell membrane is an ideal cell-mediated drug delivery platform with inherent biocompatibility, but lacks of specific targeting properties to increase drug accumulation in the psoriatic site.

Article Synopsis
  • Organ transplantation often faces challenges with T-cell-mediated rejection, which triggers a strong immune response against the graft.
  • Long-term use of immunosuppressive drugs like rapamycin can have severe side effects, so researchers are exploring alternative methods, such as using PD-L1 nanovesicles (PD-L1 NVs) to create a more balanced immune response.
  • The study shows that combining low doses of rapamycin with PD-L1 NVs not only suppresses T-cell activation but also enhances allograft tolerance and promotes the induction of regulatory T cells, making it a promising approach for improving organ transplant outcomes.
