Vulnerable atherosclerotic plaques serve as the primary pathological basis for fatal cardiovascular and cerebrovascular diseases. The precise identification and treatment of these vulnerable plaques hold paramount clinical importance in mitigating the incidence of myocardial infarction and stroke. Nevertheless, the identification of vulnerable plaques within the diffuse atherosclerotic plaques dispersed throughout the systemic circulation continues to pose a substantial challenge in clinical practice.

Atherosclerosis is a typical chronic inflammatory vascular disease that seriously endangers human health. At present, oral lipid-lowering or anti-inflammatory drugs are clinically used to inhibit the development of atherosclerosis. However, traditional oral drug treatments have problems such as low utilization, slow response, and serious side effects.

This study aims to optimize the conditions for the formation of neutrophil extracellular traps(NETs) in vitro, so as to establish a relatively stable experimental research platform. Different conditions were compared, including commonly used laboratory animals(rats and mice) and a variety of cell sources(bone marrow neutrophils and peripheral blood neutrophils separated by percoll density gradient centrifugation). Different inducers like lipopolysaccharide(LPS) and phorbol 12-myristate 13-acetate(PMA) were used for induction in vitro.

Increasing research has shown that the abnormal expression of circRNAs is closely related to tumorigenesis, apoptosis, and patient prognosis in cervical cancer. This study aimed to reveal the procancer role of circIL21R in cervical cancer and investigate its related molecular mechanisms. Bioinformatics analysis revealed that circIL21R promotes the progression of cervical cancer via the miR-1205/PTBP1 axis.

Res Pract Thromb Haemost

August 2022

State Key Laboratory of Experimental Hematology, National Clinical Research Center for Blood Diseases, Institute of Hematology & Blood Diseases Hospital, Chinese Academy of Medical Sciences & Peking Union Medical College, Tianjin Laboratory of Blood Disease Gene Therapy, CAMS Key Laboratory of Gene Therapy for Blood Diseases, CAMS Center for Stem Cell Medicine, PUMC Department of Stem Cell and Regenerative Medicine Tianjin China.

Article Synopsis
  • The study focuses on identifying risk factors for the development of factor VIII (FVIII) inhibitors in hemophilia A patients through advanced genomic techniques.
  • RNA-sequencing and whole-genome bisulfite sequencing were performed on blood samples from patients with and without inhibitors, revealing significant differences in gene expression related to immune response.
  • Results indicated an upregulation of immune activation genes, particularly those linked to T-cell activation, suggesting that lncRNA and methylation changes could be key factors in inhibitor production and may lead to new treatment strategies.

Micro(mi)RNAs play a critical regulatory role in the progression and metastasis of pancreatic cancer (PC). In this study, we aimed to reveal the mechanisms of miR-374b-5p in regulating epithelial-mesenchymal transition (EMT) in PC. Gene Expression Omnibus datasets (GSE24279 and GSE71533) and the pancreatic ductal adenocarcinoma (PDAC) cohort of The Cancer Genome Atlas were employed to screen for potential prognostic miRNAs.

Although inflammation and emphysema in patients with chronic obstructive pulmonary disease (COPD) can be ameliorated by antibiotics such as erythromycin, the impact of drug resistance is still controversial. We aimed to evaluate the role of F528, a new macrolide derivative without antibacterial effect, in cigarette smoke (CS)-induced pulmonary inflammation and emphysema in a mouse model, as well as in a macrophage cell line. The inflammatory cell number and cell type in the BALF were counted, and the levels of cytokines in the BALF and cultured cell medium were measured by ELISA.

Summary of animal models of myelodysplastic syndrome.

Animal Model Exp Med

March 2021

NHC Key Laboratory of Human Disease Comparative Medicine Beijing China.

Myelodysplastic syndrome (MDS) is a malignant tumor of the hematological system characterized by long-term, progressive refractory hemocytopenia. In addition, the risk of leukemia is high, and once it develops, the course of acute leukemia is short with poor curative effect. Animal models are powerful tools for studying human diseases and are highly effective preclinical platforms.

Two phloroglucinol compounds(1-2) were isolated and purified from 95% ethanol extract of Dryopteris fragrans through various column chromatographies on silica gel, Sephadex LH-20, medium pressure column chromatography, and preparative HPLC. Their structures were elucidated as 2',4',6'-trihydroxy-5'-methyl acetate-3'-methyl-1'-butyrophenone(1) and aspidinol B(2) based on their chemical and physicochemical methods and spectroscopic data. Compound 1 is a new phloroglucinol compound named "dryofraginol".

Chest computed tomography (CT) scans of coronavirus 2019 (COVID-19) disease usually come from multiple datasets gathered from different medical centers, and these images are sampled using different acquisition protocols. While integrating multicenter datasets increases sample size, it suffers from inter-center heterogeneity. To address this issue, we propose an augmented multicenter graph convolutional network (AM-GCN) to diagnose COVID-19 with steps as follows.

Article Synopsis
  • Immune tolerance induction (ITI) therapy for hemophilia A is currently too expensive in China, making it difficult to manage children with high-titer inhibitors.
  • A pilot study was conducted on children with poor-risk status, utilizing a low-dose plasma-derived factor VIII/von Willebrand factor concentrate, sometimes combined with immunosuppressants like rituximab and prednisone.
  • The study found that 87.5% of participants responded positively to the treatment, reducing bleeding episodes significantly and proving to be cost-effective compared to traditional high-dose regimens, although further research is necessary.
