Tumor-associated macrophages (TAMs) undergo metabolic reprogramming, encompassing glucose, amino acid, fatty acid metabolism, tricarboxylic acid (TCA) cycle, purine metabolism, and autophagy, within the tumor microenvironment (TME). The metabolic interdependencies between TAMs and tumor cells critically influence macrophage recruitment, differentiation, M2 polarization, and secretion of epithelial-mesenchymal transition (EMT)-related factors, thereby activating intratumoral EMT pathways and enhancing tumor cell invasion and metastasis. Tumor cell metabolic alterations, including hypoxia, metabolite secretion, aerobic metabolism, and autophagy, affect the TME's metabolic landscape, driving macrophage recruitment, differentiation, M2 polarization, and metabolic reprogramming, ultimately facilitating EMT, invasion, and metastasis.

Background: There are different types of vena cava filter (VCF) available in clinical practice. However, limited data exist to determine whether one type is superior to another, and no single VCF is universally recommended in clinical guidelines. The objective of this study was to investigate the safety and efficacy of a novel VCF, Octoparms, for the prevention of pulmonary embolism (PE) and to compare it with the Celect filter.

Being activated by deacetylation, peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) has become an important regulator of metabolic-related diseases. The activation of Sirtuin 1 (Sirt1) by resveratrol was likely to deacetylate PGC-1α. However, the role of deacetylated PGC-1α in the alleviation of activated Sirt1 on type 2 diabetes mellitus (T2DM)-related fatty liver disease (FLD) remained unexplored.

  • - Metabolic memory (MM) contributes to delayed wound healing in diabetics, and this study explores how transient hyperglycemia damages vascular endothelial cells by reducing METTL3 expression, affecting RNA methylation and cellular metabolism.
  • - The application of Nocardia rubrum cell wall skeleton (Nr-CWS) boosts METTL3 levels, aiding in the recovery of endothelial cell function that is compromised by transient hyperglycemia.
  • - The research highlights METTL3's crucial role in diabetic wound healing, while suggesting that Nr-CWS could be a promising new treatment strategy for improving recovery in diabetic patients.

  • The study investigates how therapeutic hypothermia (TH) affects heart damage caused by ischemia-reperfusion (I/R) injury using rat models and cardiac cells.
  • Results show that TH decreases the size of heart damage, lowers serum levels of injury markers, and reduces inflammation and cell death associated with I/R injury.
  • TH's protective effects may be linked to the DKK3/Wnt/β-catenin signaling pathway, suggesting potential mechanisms for treating heart conditions.

Aims: The emergence of drug-resistant tuberculosis has necessitated novel treatments like the pretomanid, bedaquiline and linezolid (BPaL) regimen. This study investigated the association of drug-induced liver injury (DILI) with the BPaL regimen compared to first-line antituberculosis drugs (isoniazid, rifampin, pyrazinamide and ethambutol [HRZE]).

Methods: A retrospective pharmacovigilance analysis was conducted using data from the US Food and Drug Administration Adverse Event Reporting System database from July 2019 to June 2023.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by the abnormal proliferation of fibroblast-like synoviocytes and changes in the joint synovium, including elevated reactive oxygen species, decreased pH, and reduced oxygen content. In this study, we synthesized a novel nanocomposite material, namely HA-PBA-TiO Janus nanocomposite, by in situ etching in prussian blue analogs doped with Co and Ni, followed by the growth of TiO nano-flowers and encapsulation in hyaluronic acid. When these janus nanoparticles diffused to the inflammatory sites of RA, they exhibited outstanding photocatalytic water-splitting ability under 660 nm laser irradiation, generating H and O.

Purpose: Because only a subset of cancer patients can benefit from immunotherapy, identifying predictive biomarkers of ICI therapy response is of utmost importance.

Methods: We analyzed the association between hemoglobin (HGB) levels and clinical outcomes in 1,479 ICIs-treated patients across 16 cancer types. We explored the dose-dependent associations between HGB levels and survival and immunotherapy response using the spline-based cox regression analysis.

  • The study examined risk factors for kidney issues in children with immunoglobulin A (IgA) vasculitis, focusing on a group of 117 patients aged 16 and under.
  • Among the patients, 41.9% developed renal injury, with factors like age, gastrointestinal bleeding, and certain blood markers linked to this outcome.
  • The final analysis identified older age, elevated D-dimer levels, and gastrointestinal bleeding as significant independent risk factors for kidney damage in these children.

Wingless-type inducible signaling pathway protein-1 (WISP1) is a newly recognized adipokine, associated with obesity and type 2 diabetes (T2DM). This study aimed to investigate the effect of metabolic and bariatric surgery (MBS) on WISP1 circulating (serum) levels and tissue expression using rat models. We initially investigated whether WISP1 circulating levels were altered between the T2DM and normal rats.

CypA/TAF15/STAT5A/miR-514a-3p feedback loop drives ovarian cancer metastasis.

Oncogene

November 2024

Xuzhou Key Laboratory of Laboratory Diagnostics, Xuzhou Medical University, Xuzhou City, Jiangsu Province, China.

Article Synopsis
  • Cyclophilin A (CypA) is identified as a key driver of epithelial-mesenchymal transition (EMT) in ovarian cancer, correlating higher expression with poor patient prognosis.

  • CAR-T therapy has improved treatment options for relapsed/refractory B-cell lymphoblastic leukemia, but it can cause serious side effects like cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS).
  • A study involving 93 patients showed that while most (81.7%) experienced CRS and a few (5.3%) experienced ICANS, these conditions did not significantly impact treatment effectiveness or patient survival rates.
  • Ultimately, the findings suggest that experiencing CRS or ICANS does not lead to worse outcomes for patients receiving anti-CD19 CAR-T therapy.

Endobronchial lipoma (EL) is a rare benign lung tumor, and its incidence rate only accounts for a tiny proportion of all lung tumors. EL has non-specific clinical symptoms and signs, and chest computed tomography is helpful in diagnosis. When fat density nodules are found in the bronchial lumen with no enhancement, EL should be considered.
