Background: Extraskeletal Ewing's sarcoma (EES) is a rare tumor, and there is currently no predictive model for overall survival of EES patients. This study sought to use data from the Surveillance, Epidemiology, and End Results (SEER) database to develop a clinical predictive model that could be used to assess the prognosis of EES patients.
Methods: We selected and downloaded prognostic data on 356 patients diagnosed with extraskeletal Ewing's sarcoma based on screening criteria, These patients were distributed between 2004 and 2015.

Lung adenocarcinoma (LUAD) represents one of the most common subtypes of lung cancer with high rates of incidence and mortality, which contributes to substantial health and economic demand across the globe. Treatment today mainly consists of surgery, radiotherapy, and chemotherapy, but their efficacy in advanced stages is often suboptimal and emphasizes the clear need for new biomarkers and therapeutic targets. In this study, single-cell RNA sequencing (scRNA-seq), spatial transcriptomics (ST), bulk RNA-seq, multiplexed quantitative immunofluorescence (QIF), and in vivo and in vitro experiments were performed to determine the heterogeneity, spatial distribution, and clinical significance of CAFs in chordoma. ScRNA-seq was performed on 87,693 single cells derived from seven tumor samples and four control nucleus pulposus samples.

Immunotherapy resistance in bladder cancer (BLCA) is associated with elevated levels of sialic acid-binding immunoglobulin-like lectin (Siglec15). This protein plays a crucial role in fostering a noninflammatory tumor microenvironment (TME), which is conducive to cancer progression. Our study confirmed that the overexpression of Siglec15 led to a reduction in CD8 T cell infiltration.

Innate immunity recognizes tumor derived damage-associated molecular patterns (DAMPs) and initiates the anti-tumor adaptive responses. The asymmetric [5 + 2] cycloaddition of VECs remains to be comparatively rare. Herein, we reported an enantioselective formal [5 + 2] annulation of 3-hydroxyquinolinones and vinylethylene carbonates (VECs) through Pd- and Cu-catalyzed tandem allylation/asymmetric [1,3]-rearrangement/hemiketalization sequences. The strategy exhibits good substrate tolerance, affording a wide range of tricyclic quinolinones bearing two adjacent quaternary stereocenters in moderate to good yields with excellent enantioselectivities.

Clinical data were compared between initial (I-SRNS) and secondary (S-SRNS) steroid resistance subgroups and Cox regression analysis was used to determine risk factors for chronic kidney disease (CKD) and CKD stage 5 (CKD5) in patients with SRNS.
