Disease diagnosis, prognosis, and therapy choice progressively rely on good-quality Deoxyribonucleic acid (DNA) for molecular analysis. As tissue processing is routinely performed worldwide with ancient techniques using toxic and DNA-damaging formaldehyde, the quality of DNA isolated from embedded tissues used for diagnostics is poor. We used a novel formalin-free tissue embedding method to process tissues and show that, after 5 years, DNA quality is superior to formalin fixation.

Current methods for the macrocyclization of phage-displayed peptides often rely on small molecule linkers that nonspecifically react with targeted amino acid residues. To expand tool kits for more regioselective macrocyclization of phage-displayed peptides, this study explores the unique condensation reaction between an N-terminal cysteine and nitrile along with the reactivity of an internal cysteine. Five 2-cyanopyrimidine derivatives were synthesized for this purpose and evaluated for their selective macrocyclization of a protein-fused model peptide.

Sensor networks generate vast amounts of data in real-time, which challenges existing predictive maintenance frameworks due to high latency, energy consumption, and bandwidth requirements. This research addresses these limitations by proposing an edge-cloud hybrid framework, leveraging edge devices for immediate anomaly detection and cloud servers for in-depth failure prediction. A K-Nearest Neighbors (KNNs) model is deployed on edge devices to detect anomalies in real-time, reducing the need for continuous data transfer to the cloud.

Inflammatory bowel disease (IBD) is a complex, multisystemic disease and is associated with ocular pathology in 4-12% of patients. In general, ocular disease affects Crohn's patients more frequently than those with ulcerative colitis. Episcleritis and uveitis are the most common presentations, with episcleritis often correlating with IBD flares, whereas uveitis presents independently of IBD activity and, in some cases, may even alert clinicians to a new diagnosis of IBD.

: The high recurrence rate of bacterial vaginosis (BV) after antibiotic treatment is at least partially attributed to resistant bacteria. The CAPRISA 083 (CAP083) study investigated the influence of metronidazole (MTZ) treatment on the vaginal microbiome in 56 South African women diagnosed with BV. To explore the etiology of recurrent BV in this cohort, we retrospectively analyzed vaginal swabs collected in CAP083 before and after MTZ treatment.
