Article Synopsis
  • Migraine, particularly chronic migraine (CM), is a complex neurological disorder with poorly understood causes, involving activation of specific brain regions like the insula and amygdala in response to pain.
  • The study used nitroglycerin (NTG) to create a mouse model of chronic migraine and investigated the effects of a treatment called Olcegepant (OLC) on pain sensitivity and brain activity.
  • Results indicated that OLC improved pain thresholds and reduced anxiety-like behaviors in the mice, with whole-brain imaging revealing significant activation in areas like the isocortex, somatomotor, and somatosensory regions, suggesting these as potential targets for future migraine treatments.

CD7 targeted CAR-T has demonstrated potential in the treatment of T cell malignancies but no study has been reported about its potential in the prophylaxis of GVHD in allo-HSCT. Here we reported a special case that a boy diagnosed with refractory acute T lymphoblastic leukemia (T-ALL) was treated with universal CD7 targeted CAR-T (CD7 UCAR-T) and parent-derived peripheral blood stem cells (PBSCs). Complete remission and full engraftment of donor was observed.

T-cell acute lymphoblastic leukemia (T-ALL) represents an area of highly unmet medical needs. Once relapsed, patients have limited treatment options and poor prognosis. T-ALL antigens such as CD7 is extensively expressed in normal T cells and natural killer (NK) cells, and extending the success of CAR-T therapy to T cell malignancies was challenged by CAR-T cell fratricide, high production cost, and potential product contaminations.

Background: Recently, increasing evidence has demonstrated that IL-10 single nucleotide polymorphisms (SNPs) are associated with the risk of acute leukemia (AL), but the findings of different articles remain controversial. Thus, we performed a meta-analysis to further investigate the exact roles of IL-10 SNPs in AL susceptibility.

Methods: Six common Chinese and English databases were utilized to retrieve eligible studies.

CD19-targeted chimeric antigen receptor T lymphocytes (CAR-T) has demonstrated a high proportion of complete remission in the treatment of relapsed refractory acute B cell lymphoblastic leukemia (r/r B-ALL). It is of great clinical significance to explore which factors will impact long-term disease-free survival of patients with r/r B-ALL after CAR-T therapy without bridging bone marrow transplantation. Our study found that, in patients with r/r B-ALL without bridging transplantation, the patients' age; infusion dosage; whether they had undergone allo-stem cell transplantation before CAR-T therapy, using CD-19-targeted or CD19/CD22-dual-targeted CAR-T; whether there is fusion gene; tumor burden before therapy; and comorbidity had no significant relationship with their long-term disease-free survival.

Article Synopsis
  • The anterolateral thigh flap is favored for its minimal damage to donor sites, preservation of major blood vessels, and ample soft tissue, although it is less commonly used in children due to their smaller blood vessels and rapid physiological changes.
  • The study involved 26 pediatric patients aged 2 to 13 with significant soft tissue defects mostly in the lower limbs, where injuries were caused by various types of trauma.
  • Despite some complications like vascular crises and infections, the free anterolateral thigh flap proved to be a safe and effective method for reconstructing soft tissue defects in children, though scar hyperplasia was noted in a number of cases.
