They bind SF3B1 and PHF5A, components of the human spliceosome, and they alter pre-mRNA splicing. Detailed analysis of the active site led us to investigate a narrow pocket within the binding site that surrounds the α,β-unsaturated amide portion of meayamycin.

The cumulated π system of a nonsymmetric allene contains three distinct unsaturated carbons that imbue it with unique reactivity toward radicals as compared to its alkene and alkyne counterparts. Despite the synthetic potential of these versatile building blocks, electrochemical transformations of allenes have been historically underexplored. In previous work, a Large Eddy Simulation turbulence (LES) algorithm was developed for finite element-based computational fluid dynamics, using a hybrid continuous-discontinuous Galerkin scheme. The test case for this was a backward-facing step, which is a well-known example with published experimental results for validation. Uncontrolled degradation and collapse of stalled replication forks (RFs) are primary sources of genomic instability, yet the molecular mechanisms for protecting forks from degradation/collapse remain to be fully elaborated. Here, we show that polynucleotide kinase-phosphatase (PNKP) localizes at stalled forks and protects stalled forks from excessive degradation. The loss of PNKP results in nucleolytic degradation of nascent DNA at stalled RFs.

A novel Gram-stain-positive, black-pigmented bacterium, designated as WL48A , was isolated from the surface of badland sedimentary rock in the Red Desert of Wyoming and characterized using a polyphasic taxonomic approach. Good growth occurred at 28-32 °C, pH 7-9, and NaCl less than 1% (w/v). Colonies, growing well on International Streptomyces Project media (ISP) 3 and ISP 7, were black and adhering to the agar.

Oxetanes are valuable motifs in medicinal chemistry applications, with demonstrated potential to serve as bioisosteres for an array of functional groups. Through the visible-light-mediated photoredox hydrodecarboxylation of 2-aryl oxetane 2-carboxylic acids this work enables access to the products of a [2+2]-photocycloaddition between alkenes and aryl aldehydes without the challenges associated with a traditional UV-light-mediated Paternò-Büchi reaction. Investigation into the hydrodecarboxylation mechanism reveals substrate-dependent modes of initiation under the conditions reported herein.

The interlamellar matrix (ILM), located between the annular layers of the intervertebral disc (IVD), is an adhesive component which acts to resist delamination. Investigating the mechanical properties of the ILM can provide us with valuable information regarding risk of disc injury; however given its viscoelastic nature, it may be necessary to conduct preconditioning on tissue samples before measuring these ILM properties. Therefore, the aim of this study was to optimize mechanical testing protocols of the ILM by examining the effect of preconditioning on stiffness and strength of this adhesive matrix.

Barium zirconium trisulfide (BZS) is a three-dimensional (3D) perovskite with optoelectronic properties suitable for photovoltaic (PV) and light-emitting diode (LED) applications that is conventionally reported in the orthorhombic (62) symmetry. Synchrotron X-ray diffraction, thermal analysis, and Raman and absorption spectroscopy revealed three high-temperature polymorphs that appear when BZS is heated in air prior to complete oxidation (BaZrS + 5O → BaSO + ZrO + 2SO↑) at 700 °C with the approximate stability ranges: BaZrS IV (62)T < 400 °CBaZrS III (63)400 °C ≤ T ≤ 500 °CBaZrS II14/ (140)500 °C ≤ T ≤ 700 °CDifferential scanning calorimetry (DSC) revealed exothermic features accompanying the IV → III and III → II phase changes.
