Département de Chimie, Institut Courtois, Quebec Center for Advanced Materials, Regroupement Québécois sur les Matériaux de Pointe, Université de Montréal, C.P. 6128 Succ. Centre-ville, Montréal H3C 3J7, Québec, Canada.

A surface-enhanced Raman scattering (SERS) biosensor with minimal invasiveness and high spatial resolution has been developed as a nanoendoscope to detect changes in protein concentrations at specific sites in biological tissues. While generally applicable to various tissues or proteins, the SERS nanoendoscope is demonstrated for the quantitative detection of S100β, an astrocytic protein whose plasmatic levels are known to vary in several neuropathologies such as Alzheimer's disease, schizophrenia, Down syndrome, Parkinson's disease and epilepsy, but for which intratissular levels have not been locally monitored, demonstrating key attributes of the SERS nanoendoscope. The SERS nanoendoscope is fabricated with densely and well-dispersed deposited gold nanoparticles modified with anti-S100β primary antibody on pulled optical fibers with a tip diameter of 700 nm, conducive to noninvasive and regiospecific detection of the S100β protein in different regions of mouse brain slices under different physiological stimuli with micrometer resolution.

Adsorption and Permeation Events in Molecular Diffusion.

Molecules

October 2024

CNRS - Université de Montréal CRM-CNRS, 6128 Succ Centre-Ville, Montréal, QC H3C 3J7, Canada.

How many times can a diffusing molecule permeate across a membrane or be adsorbed on a substrate? We employ an encounter-based approach to find the statistics of adsorption or permeation events for molecular diffusion in a general confining medium. Various features of these statistics are illustrated for two practically relevant cases: a flat boundary and a spherical confinement. Some applications of these fundamental results are discussed.

Article Synopsis
  • Understanding the dietary habits of First Nations is essential for tackling health disparities, with history affecting their food practices and health for generations.
  • Indigenous peoples on reserves are not included in Canada’s dietary surveys, prompting a study that evaluates First Nations adults' diet quality using HEFI-2019 and C-HEI 2007 across ninety-two communities.
  • Results showed low adherence to dietary guidelines (HEFI-2019 score of 35/80 and C-HEI 2007 score of 49/100), with factors like age, education, and traditional food intake significantly impacting diet quality.

Background: More and more people suffer from concomitant chronic physical diseases and common mental disorders, calling for integrated self-management support in primary care. However, self-management support of chronic physical diseases and common mental disorders is not clearly operationalized by guidelines and is still conducted in silos by primary care nurses, especially in favour of chronic diseases. This study aims to better understand primary care nurses' experience of integrated self-management support for people with physical chronic diseases and common mental disorders.

Modeling the impact of heat stress on the toxicokinetics of toluene and acetone.

Arch Toxicol

February 2024

Centre de recherche en santé publique, Université de Montréal, C.P. 6128 Succ. Centre-ville, Montreal, (Qc.), H3C 3J7, Canada.

Many workers can be exposed simultaneously to heat and volatile chemicals. In a controlled human exposure study, it was observed that an increase in ambient temperature was associated with increased blood concentrations for acetone and toluene. Based on the expected changes in physiological parameters that occur with an increase in ambient temperature, we aimed to develop a PBPK model for acetone and toluene that could account for the impact of temperature on the kinetics of these solvents.

Article Synopsis
  • Bovine serum albumin (BSA) is commonly used in biosensing as a blocking buffer, but human serum albumin (HSA) has a significant impact on binding assays and has been underexplored.
  • A study utilized surface plasmon resonance (SPR) to investigate the effects of different albumins and human serum on a simple binding assay involving human IgG and goat anti-human IgG, revealing that HSA substantially influences the SPR shifts and binding constants.
  • The findings suggest BSA is effective for surface blocking but recommend using HSA for optimizing assays intended for clinical applications involving human blood or serum, highlighting the necessity of considering specific protein interactions in such contexts.

Optoplasmonic Effects in Highly Curved Surfaces for Catalysis, Photothermal Heating, and SERS.

ACS Appl Mater Interfaces

October 2023

Department of Material Science and Metallurgy, University of Cambridge, 27 Charles Babbage Road, Cambridge, U.K. CB3 0FS.

Surface curvature can be used to focus light and alter optical processes. Here, we show that curved surfaces (spheres, cylinders, and cones) with a radius of around 5 μm lead to maximal optoplasmonic properties including surface-enhanced Raman scattering (SERS), photocatalysis, and photothermal processes. Glass microspheres, microfibers, pulled fibers, and control flat substrates were functionalized with well-dispersed and dense arrays of 45 nm Au NP using polystyrene--poly-4-vinylpyridine (PS--P4VP) and chemically modified with 4-mercaptobenzoic acid (4-MBA, SERS reporter), 4-nitrobenzenethiol (4-NBT, reactive to plasmonic catalysis), or 4-fluorophenyl isocyanide (FPIC, photothermal reporter).

Detailed mechanistic investigations of the interrelated roles of multiple key structure-directing agents in the growth solution of Au nanoparticles (AuNPs) is required for the optimization of synthetic protocols. Here, we report a robust seed-mediated growth strategy for synthesizing multibranched NPs (MB-AuNPs) with monodispersed size distribution, and investigate the roles of Ag ions and 4-(2-hydroxyethyl)piperazine-1-ethanesulfonic acid (HEPES) based on an overgrowth synthesis approach. The intertwining roles of Ag , surface-capping stabilizers, and reducing agents were elucidated, and used to control the morphology of MB-AuNPs.

Département de Chimie, Quebec Center for Advanced Materials, Regroupement Québécois Sur Les Matériaux de Pointe, and Centre Interdisciplinaire de Recherche Sur Le Cerveau Et L'apprentissage, Université de Montréal, CP. 6128 Succ. Centre-Ville, Montréal, QC, H3C 3J7, Canada.

A series of thermally shrinkable polymer surface-enhanced Raman scattering (SERS) substrates were prepared with bimetallic Au and Ag (oxidized or not) films and with Au nanoparticles (AuNPs) located at different places in the layered structure to evaluate the synergistic effect of different known SERS amplification methods to enhance the Raman signal for low concentration dopamine detection. A bimetallic Au and Ag layered structure improved the Raman signal by 5 and 2 times compared to the single-layered Au and Ag films. Oxidizing the Ag layer prior to deposition of Au further improved the signal by a factor of 2, while adding AuNP on wrinkled films increased another 10 times the intensity of the Raman signal.

In Nunavik (Northern Quebec, Canada), some mining projects are envisioned, that could increase the contamination of the environment by various chemicals, including rare earth elements (REEs), and implicitly Inuit population exposure. The objective of this study was to determine the baseline biological exposure of the population to these elements, before the potential mining development occurs. In the framework of the 2017 Qanuilirpitaa? Inuit health survey, urine samples were obtained from a representative sample of the adult Nunavik population, which were used to constitute 30 pooled samples according to age, sex and Nunavik subregions.
