Models of Classroom Assessment for Course-Based Research Experiences.

Course-based research pedagogy involves positioning students as contributors to authentic research projects as part of an engaging educational experience that promotes their learning and persistence in science. To develop a model for assessing and grading students engaged in this type of learning experience, the assessment aims and practices of a community of experienced course-based research instructors were collected and analyzed. This approach defines four aims of course-based research assessment - 1) Assessing Laboratory Work and Scientific Thinking; 2) Evaluating Mastery of Concepts, Quantitative Thinking and Skills; 3) Appraising Forms of Scientific Communication; and 4) Metacognition of Learning - along with a set of practices for each aim.

Pharmaceutical companies use the quality by design (QbD) approach to build high-quality drug products. A thorough understanding of risk factors is required to successfully employ QbD. In order to better understand risk factors that potentially impact drug product quality and inform future QbD approaches, we hypothesized root causes of drug product recalls based on publicly available data and a retroactive analysis of drug products recalled by the United States Food and Drug Administration (USFDA) from 2012 to 2018.

Characterization of covalent crosslinking strategies for synthesizing DNA-based bioconjugates.

An adapted strategy from the conventional 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) crosslinking method was developed to form a covalently coupled phosphoramidated single stranded DNA (ssDNA). Matrix assisted laser desorption ionization-time of flight (MALDI-TOF) results demonstrated that the phosphoramidated ssDNA conjugate is stable for several days, and that phosphoramidation occurred exclusively at the 5' phosphate of ssDNA. A reversed phase high-performance liquid chromatography (RP-HPLC) method with UV detection was developed to determine the yield of conjugates.

In this paper, spatial and temporal variations of trihalomethane (THM) concentrations were analyzed including chloroform trichloromethane (TCM), bromodichloromethane (BDCM), dibromochloromethane (DBCM), and tribromomethane (TBM) in Yancheng City in Jiangsu Province, China. The water samples were collected monthly from January 2014 to January 2017 from four tap water sampling sites (S1, S2, S3, and S4) and two finished water sampling sites (WTP1 and WTP2) for THM analysis. The results showed that the mean concentrations during the study period for TCM, BDCM, DBCM, and TBM were 7, 15.

This study investigated potential changes in flow, total suspended solid (TSS) and nutrient (nitrogen and phosphorous) loadings under future climate change, land use/cover (LULC) change and combined change scenarios in the Wolf Bay watershed, southern Alabama, USA. Urbanization increases nutrient loading and lowers residence times for processing of reactive solutes, including nitrate, total dissolved nitrogen, orthophosphate, and dissolved organic carbon), which leads to increased stream concentrations and mass export. Stormwater control measures mitigate the impacts of urbanization, and have the potential to improve stream water quality, however the net effect instream is not well understood. We monitored two urban and two suburban watersheds in Charlotte, NC to determine if mitigation controlled the fraction of total mass export during storm, if development classification as either urban or suburban (defined by the age, density and distribution of urban development) controlled storm nutrient and carbon dynamics, and if stormwater control measures were able to change stream water chemistry.

The detailed mechanism for DNA methylation homeostasis relies on an intricate regulatory network with a possible contribution from methyl-CpG-binding domain protein 3 (MBD3). The coffee strip-picking harvesting method, preferred in Brazil, results in high percentages of immature and overripe beans, as the fruits in a single tree branch do not reach ripeness at the same time. This practice, together with inappropriate processing and storage conditions, contribute to the production of high amounts of defective coffee beans in Brazil, which upon roasting will impart negative sensory aspects to the beverage. Therefore, the development of analytical methodologies that will enable the discrimination and quantification of defective and non-defective coffees after roasting is rather desirable.

The quality of the coffee beverage is negatively affected by the presence of defective coffee beans and its evaluation still relies on highly subjective sensory panels. To tackle the problem of subjectivity, sophisticated analytical techniques have been developed and have been shown capable of discriminating defective from non-defective coffees after roasting. However, these techniques are not adequate for routine analysis, for they are laborious (sample preparation) and time consuming, and reliable, simpler and faster techniques need to be developed for such purpose.

Differentiation of cancer cells in two-dimensional and three-dimensional breast cancer models by Raman spectroscopy.

We demonstrate the first application of Raman spectroscopy in diagnosing nonmalignant, premalignant, malignant, and metastatic stages of breast cancer in a three-dimensional (3-D) cell culture model that closely mimics an in vivo environment. Comprehensive study comparing classification in two-dimensional (2-D) and 3-D cell models was performed using statistical methods composed of principal component analysis for exploratory analysis and outlier removal, partial least squares discriminant analysis, and elastic net regularized regression for classification. Our results show that Raman spectroscopy with an appropriate classification tool has excellent resolution to discriminate the four stages of breast cancer progression, with a near 100% accuracy for both 2-D and 3-D cell models.

We report a label-free fluorescence lifetime profiling strategy to classify breast cancer cells, MCF10CA1h (malignant), MCF10A (nonmalignant), and MCF10AneoT (premalignant) in different stages of malignancy. Fluorescence Lifetime Imaging Microscopy (FLIM) was used to record the lifetime of autofluorescence of endogenous flavin in MCF10 cells in different stages of malignancy. Predominant differences in lifetimes ascertained by multi-exponential fitting curves can be attributed to the different forms of flavin protein; flavin mononucleotide (FMN), free flavin adenine dinucleotide (FAD), semiquinone, and bound FAD.

Folic acid protected silver nanocarriers for targeted drug delivery.

A one-step approach towards the synthesis of folic acid (FA) coated silver nanoparticles (AgNPs) and doxorubicin (DOX) for drug delivery is proposed. The FA-AgNPs were 23 +/- 2 nm in diameter and showed good monodispersity with a characteristic surface plasmon peak at 409 nm. Taking advantage of the enhanced Raman signal of FA on AgNPs and targeting specificity of FA to folate-receptor expressing cancer cells, the as-prepared FA-AgNPs showed excellent receptor-mediated cellular uptake, as demonstrated both by surface enhanced raman scattering (SERS) imaging and fluorescent lifetime imaging (FLIM).
