The Search for Disease-Modifying Therapies in Pulmonary Hypertension.

J Cardiovasc Pharmacol Ther

July 2019

1 Division of Cardiology, Department of Medicine, Center for Pulmonary Vascular Biology and Medicine, Pittsburgh Heart, Lung, Blood, and Vascular Medicine Institute, University of Pittsburgh School of Medicine, Pittsburgh, PA, USA.

Pulmonary hypertension (PH) and its severe subtype pulmonary arterial hypertension (PAH) encompass a set of multifactorial diseases defined by sustained elevation of pulmonary arterial pressure and pulmonary vascular resistance leading to right ventricular failure and subsequent death. Pulmonary hypertension is characterized by vascular remodeling in association with smooth muscle cell proliferation of the arterioles, medial thickening, and plexiform lesion formation. Despite our recent advances in understanding its pathogenesis and related therapeutic discoveries, PH still remains a progressive disease without a cure.

Objective: Serious games are a growing form of psychoeducation, although few studies have evaluated serious games for patients with advanced cancer. The purpose of this study was to develop and assess the initial acceptability of a serious game to teach women with advanced cancer self-advocacy skills, including communication, decision-making, and social connectivity, to improve their quality of life with cancer.

Materials And Methods: We conducted a multistage, user-centered codesign process to develop the content of the game that was consistent with our work on how patients self-advocate and patients' preferences for the game.

Multicellular tumor spheroid (MCTS) cultures represent more physiologically relevant in vitro cell tumor models that recapitulate the microenvironments and cell-cell or cell-extracellular matrix interactions which occur in solid tumors. We characterized the morphologies, viability, and growth behaviors of MCTSs produced by 11 different head and neck squamous cell carcinoma (HNSCC) cell lines seeded into and cultured in ultra-low attachment microtiter plates (ULA-plates) over extended periods of time. HNSCC MCTS cultures developed microenvironments, which resulted in differences in proliferation rates, metabolic activity, and mitochondrial functional activity between cells located in the outer layers of the MCTS and cells in the interior.

This article examines whether emotional suppression is associated with socioeconomic position (SEP) in a community sample of Black and White men, and whether emotional suppression may help explain the aggregation of multiple biopsychosocial risk factors for cardiovascular disease at lower SEP (social support, depression, cardiovascular stress reactivity). Aim 1 tests whether multiple indicators of SEP show a consistent graded association with self-reported trait suppression, and whether suppression mediates associations between SEP and perceived social support and depressive affect. Aim 2 tests whether suppression during a laboratory anger recall task mediates associations between SEP and cardiovascular reactivity to the task.

Animal and clinical studies demonstrate that cancer drug combinations (DCs) are more effective than single agents. However, it is difficult to predict which DCs will be more efficacious than individual drugs. Systematic DC high-throughput screening (HTS) of 100 approved drugs in the National Cancer Institute's panel of 60 cancer cell lines (NCI-60) produced data to help select DCs for further consideration.

The present study is an analysis of in-depth interviews with college women reporting a mental health disability and at least one experience of intimate partner violence (IPV) or sexual violence (SV) to elucidate how alcohol use is associated with both violence victimization and mental health symptoms. Our findings underscore salient alcohol-related themes in college women with histories of IPV/SV and mental health disability: alcohol use in their family of origin and/or with intimate partners, partying and heavy drinking as a normal college social context, abusive partners and SV perpetrators using alcohol as a mechanism for control and targeted rape, and worsening mental health symptoms after violence exposure, which prompted alcohol use to cope and was associated with vulnerability to more violence.

Fundamental movement skills (FMS) play a crucial role in a child's motor skill development, as their mastery is related to the acquisition and mastery of specialized sport skills and to a child's general health through the prevention of weight gain and increased level of physical activity. Thus, the accurate and reliable measurement of FMS is of related importance. This article presents the results of two studies intended to assess expert-rater agreement and inter-/intrarater scoring reliability in the administration of a new observational FMS instrument, the Furtado-Gallagher Computerized Observational Movement Pattern Assessment System (FG-COMPASS).
