An ideal codrug design should be able to control drug release, offer selectivity during drug delivery, and break down into non-toxic fragments after biodegradation. Our design incorporates d-ribofuranose as the core, with carbamate and carbonate groups as linking joints, a phosphodiester moiety as an enzyme recognition site, and lenalidomide and paclitaxel as the constituent drugs. The codrug synthesis involves seven steps with a 33% overall yield.

Article Synopsis
  • Pexidartinib is a drug used for treating tenosynovial giant cell tumor (TGCT) in patients where surgery isn't an option, and this study looked at the effects of stopping and then restarting the medication.
  • It was a phase 4 global study involving patients who had benefited from pexidartinib, allowing them to either continue treatment or stop with the option to restart later, monitoring their tumor progression and quality of life.
  • Results showed that while about 54.5% of patients who stopped the drug experienced disease progression, none of those who continued treatment saw their condition worsen over a 24-month period.

Sci Rep

November 2024

Division of Cardiology, Department of Internal Medicine, Center of data science and Biostatistics, Keelung Chang Gung Memorial Hospital, Chang Gung University College of Medicine, Taoyuan, Taiwan.

Given the limited head-to-head comparison of cardiovascular and renal outcomes between liraglutide and dulaglutide, our study aimed to investigate the clinical outcomes between dulaglutide and liraglutide in a real-world setting. In this new-user design, comparative and retrospective cohort study, patients with type 2 diabetes mellitus with prescription for GLP-1RAs from January 1, 2016 to December 31, 2022 (n = 8,278) were included. Primary outcome was composite cardiovascular outcomes which was composed of cardiovascular death, non-fatal myocardial infarction, and non-fatal ischemic stroke.

Department of Chemical & Materials Engineering, National Central University, Jhong-Li, Taoyuan 320, Taiwan.

Strategies to design multifunctional interfaces for biosensors have been extensively investigated to acquire optimal sensitivity, specificity, and accuracy. However, heterogeneous ingredients in clinical samples inevitably generate background signals, exposing challenges in biosensor performance. Polymer coating has been recognized as a crucial method to functionalize biointerfaces by providing tailored properties that are essential for interacting with biological systems.

Dicer, a cytoplasmic type III RNase, is essential for the maturation of microRNAs (miRNAs) and is implicated in cancer progression and chemoresistance. Our previous research demonstrated that phosphorylation of Dicer at S1016 alters miRNA maturation and glutamine metabolism, contributing to gemcitabine (GEM) resistance in pancreatic ductal adenocarcinoma (PDAC). In this study, we focused on the role of Dicer phosphorylation at S1728/S1852 in GEM-resistant PDAC cells.

Branched-chain amino acids (BCAAs) are essential for muscle protein synthesis and are widely acknowledged for mitigating sarcopenia. Oligonol (Olg), a low-molecular-weight polyphenol from , has also been found to attenuate sarcopenia by improving mitochondrial quality and positive protein turnover. This study aims to investigate the effect of Olg on BCAA-stimulated protein synthesis in sarcopenia.

Tea contains various antioxidant compounds, including polyphenols, catechins, theaflavins, theasinensins, and flavonoids. Among these, epigallocatechin gallate (EGCG) is a crucial antioxidant recognized for its potent bioactivity. This study presents the synthesis of a highly selective Cu-PyC NH-based metal-organic framework (MOF) nanozyme that exhibits catecholase-like activity to assess the antioxidant capabilities of EGCG.

Objective: Ultrahigh frequencies (UHF) have been shown to selectively suppress the sensory pathway with a rapid onset and prolonged effect compared with low frequencies. Few studies have explored the feasibility of UHF electrical stimulation in treating overactive bladder. This study aimed to investigate whether bladder overactivity could be inhibited by UHF stimulation at the L6 nerve root.

N6-methyladenosine (m6A) RNA modification orchestrates cellular epitranscriptome through tuning the homeostasis of transcript stability, translation efficiency, and the transcript affinity toward RNA-binding proteins (RBPs). An aberrant m6A deposition on RNA can lead toward oncogenic expression profile (mRNA), impaired mitochondrial metabolism (mtRNA), and translational suppression (rRNA) of tumor suppressor genes. In addition, non-coding RNAs (ncRNAs), such as X-inactive specific transcript (XIST), miRNAs, and α-ketoglutarate-centric metabolic transcripts are also regulated by the m6A epitranscriptome.

The development of opioid analgesics with reduced adverse effects is an unmet need. In a previous study, we discovered a unique combination of BPRMU191 and morphinan antagonists that produced potent antinociception with reduced adverse effects after central administration (intrathecal or intracerebroventricular). BPRMU191/naltrexone exhibits notable and pharmacological properties.
