Background: Setmelanotide, a melanocortin-4 receptor (MC4R) agonist, has been shown to reduce hunger and weight in patients aged 6 years and older with proopiomelanocortin (POMC) deficiency (including biallelic variants in proprotein convertase subtilisin/kexin type 1 [PCSK1]), leptin receptor (LEPR) deficiency, or Bardet-Biedl syndrome (BBS). No approved therapies for patients younger than 6 years old currently exist. The phase 3, open-label VENTURE trial aimed to evaluate the efficacy and safety of setmelanotide in patients aged 2-5 years with POMC or LEPR deficiency or BBS.

The effect of the treatment with an injectable product containing eprinomectin on milk yield, milk composition, and somatic cell count (SCC) was investigated in naturally infected dairy goats with low gastrointestinal nematode faecal egg and pulmonary larval counts. The study involved 394 lactating goats and was conducted on a farm in southern Spain. Before treatment, faeces from all animals were analysed to establish two homogeneous groups according to the gastrointestinal nematode egg counts (185 treated and 209 untreated).

Objective: We aimed to (1) evaluate the cardiac involvement, with a focus on myocarditis, in patients with Still disease included in the multicenter Autoinflammatory Disease Alliance (AIDA) Network Still disease registry; and (2) assess the predictive factors for myocarditis by deriving a clinical risk patient profile for this severe manifestation.
Methods: A multicenter observational study was established, in which consecutive patients with Still disease in the AIDA Network Still disease registry were characterized by cardiac involvement. Cardiac involvement was defined according to the presence of pericarditis, tamponade, myocarditis, and/or aseptic endocarditis.

Low-intensity focused ultrasound (LIFU) combined with intravenously circulating microbubbles has recently emerged as a novel approach for increasing delivery through the blood-brain barrier (BBB). This technique safely and transiently enables therapeutic agents to overcome the BBB, which typically poses a significant obstacle for treatment of brain disorders. Sclerosing mucoepidermoid carcinoma (SMEC) of the salivary glands is a rare variant of low-grade mucoepidermoid carcinoma with scanty cellular atypia characterized by marked fibrosis/sclerosis and a rich inflammatory infiltrate. Herein, we report 25 unpublished cases of SMEC, two of them with prominent eosinophilia (2/25; 8%) and three with abundant IgG4-positive plasma cells (3/25; 12%). In our series of salivary SMEC, molecular analysis using fluorescence in situ hybridization (FISH) and/or next-generation sequencing (NGS) provided evidence of MAML2 gene rearrangement in 18 cases of the 21 analyzable cases tested (86%), while this gene locus was intact in 3 cases (14%).

For the first time an electrochemical sensor based on nanomaterial-supported molecularly imprinted polymers (MIPs) is applied to the sensitive and specific determination of chloroquine phosphate (CHL). The sensor was produced using an electropolymerization (EP) approach, and it was formed on a glassy carbon electrode (GCE) using CHL as a template and 2-acrylamido-2-methyl-1-propane sulfonic acid (AMPS) and aniline (ANI) as functional monomers. Incorporating Prussian blue polyethyleneglycol-amine nanoparticles (PB@PEG-NH) in the MIP-based electrochemical sensor increased the active surface area and porosity.

Frontotemporal dementia (FTD) is one of the leading causes of young-onset dementia before age 65, typically manifesting as abnormal behavior (in behavioral variant FTD) or language impairment (in primary progressive aphasia). Although FTD affects all populations across the globe, knowledge regarding the pathophysiology and genetics derives primarily from studies conducted in North America and Western Europe. Globally, biomedical research for FTD is hindered by variable access to diagnosis, discussed in this group's earlier article, and by reduced access to expertise, funding, and infrastructure.

Two potent antioxidants, selenoneine and ergothioneine, have been determined simultaneously for the first time in the muscle of highly consumed fish and in fish-derived products. The extraction methods were optimized, and their subsequent analysis was performed by HPLC-ESI-MS/MS. Best extraction efficiency (106 ± 3 and 104 ± 5%) was attained by performing a simple aqueous extraction containing 100 mmol L ammonium acetate or ammonium formate as extractants.

Intravenous pembrolizumab 400 mg every 6 weeks was approved across tumor types based on pharmacokinetic modeling, which showed exposures consistent with previous standard dosing of 200 mg or 2 mg/kg every 3 weeks, and early results of cohort B of the phase 1 KEYNOTE-555 study. Results after ≥1 year of potential follow-up for all patients in cohort B of KEYNOTE-555 are presented. Patients aged ≥18 years with previously untreated stage III/IV melanoma received pembrolizumab 400 mg every 6 weeks for ≤18 cycles.
