Communicating scientific evidence: drugs for Alzheimer's disease as a case study.

Curr Med Res Opin

January 2025

Laboratory of Behavioral Neuroscience, National Institute on Aging, National Institutes of Health, Baltimore, MD, USA.

This paper reviews the scientific evidence on new anti-amyloid monoclonal antibodies for treating Alzheimer's disease as a case study for improving scientific evidence communication. We introduce five guidelines condensed from the biomedical evidence literature but adapted to the short format of science communication in e.g.

Data management and sharing.

Guided by the FAIR principles (Findable, Accessible, Interoperable, Reusable), responsible data sharing requires well-organized, high-quality datasets. However, researchers often struggle with implementing Data Management and Sharing Plans (DMSPs) due to lack of knowledge on how to do this, time constraints, legal, technical and financial challenges, particularly concerning data ownership and privacy. While patients support data sharing, researchers and funders may hesitate, fearing the loss of intellectual property or competitive advantage.

Incomplete reporting of adverse events in duloxetine trials: a meta-research survey of randomized controlled trials versus placebo.

J Clin Epidemiol

January 2025

Adult Psychiatry Department, Rennes University Hospital, Rennes, France; Centre for Population Neuroscience and Stratified Medicine (PONS), Department of Psychiatry and Clinical Neuroscience, Charité Universitätsmedizin Berlin, Berlin, Germany. Electronic address:

Objective: Relying on published data alone might be insufficient for meta-analyses to be reliable and trustworthy since selective outcome reporting is common, especially for adverse events. We investigated the existence of selective reporting and its potential for bias in a case study exploring adverse events of duloxetine in adults.

Study Design And Setting: We systematically searched all previous meta-analyses/pooled analyses on duloxetine published on PubMed for 7 indications approved by the American and European health authorities.

Studies with negative results are less likely to be published than others, potentially leading to publication bias. Introduced in 2000, trial registration could have participated in decreasing the proportion of unpublished studies. We assessed the proportion of negative randomized controlled trials (RCT) over the last 20 years.

Background: Chorioamnionitis is recognized as a major consequence of preterm premature rupture of membranes (PPROM), and a frequent cause of neonatal morbidity and mortality. The association between fetal heart rate (FHR) and chorioamnionitis remains unclear.

Objectives: The aim of this study was to evaluate the dynamics of FHR in a PPROM population at the approach of delivery according to the presence or absence of chorioamnionitis.

The issue of assessing the effectiveness of health technologies (drugs, devices, etc.) through observational studies is becoming increasingly important as registration and market access agencies consider them in their evaluation process. In this context, observational studies must be able to provide real demonstrations of a level of reliability comparable to those produced by the conventional randomized controlled trial (RCT) approach.

Shaping tomorrow: how the STEP training course pioneered noninvasive brain stimulation training for psychiatry in France.

Front Psychiatry

November 2024

French Society for Biological Psychiatry and Neuropsychopharmacology, STEP Section (Stimulation Transcrânienne En Psychiatrie), Saint-Germain-en-Laye, France.

Over the past three decades, non-invasive brain stimulation (NIBS) techniques have gained worldwide attention and demonstrated therapeutic potential in various medical fields, particularly psychiatry. The emergence of these novel techniques has led to an increased need for robust training programs to provide practitioners, whether clinicians or scientists, with the necessary skills and knowledge. In response, a comprehensive training curriculum for NIBS in psychiatry has been developed in France.

Purpose: Knowing the features of dental evolution and facial morphology, marked by an increase in the prevalence of agenesis and a tendency towards verticalization of the face, the main objective of this cross-sectional observational study was to assess the correlation between the occurrence of agenesis (of 3rd molars or other teeth) and facial morphology in the French population.

Methods: The study was conducted at the University Hospital of Rennes, France, from June 2022 to October 2022. Patients aged 12-18 years who underwent a global orthodontic assessment were included.

Article Synopsis
  • Micronutrient (MN) alterations in critically ill patients can lead to complications, but improving MN status may help as a supportive therapy.
  • This review, done by a specialized group, focuses on individual important MNs, such as vitamins A, B, C, D, E, as well as minerals like iron and zinc, to guide future research.
  • It emphasizes that high-dose single MN treatments are not advised; instead, patients should receive daily basal needs, with adjustments for higher requirements and treatment for deficiencies, and lists ongoing trials and future research priorities.

The advanced practice nurse (APN) has been introduced in France, following the 2016 health law and implementing decrees published in 2018. In this context, the French Society for Bone Marrow Transplantation and Cellular Therapy (SFGM-TC) has already issued guidelines regarding the allocation of APNs' new clinical competences and their collaboration with physicians. It is now providing new recommendations on the transversal activities that can be fulfilled by APNs, such as research, leadership, training and teaching.

Meta-analyses of phase I dose-finding studies: Application for the development of protein kinase inhibitors in oncology.

November 2024

Univ Rennes, CHU Rennes, Inserm, Centre d'investigation clinique de Rennes (CIC1414), Service de Pharmacologie Clinique, Institut de Recherche en Santé, Environnement et Travail (Irset), UMR S 1085, EHESP, Rennes, France.

This study aimed to assess the feasibility of applying two recent phase I meta-analyses methods to protein kinase inhibitors (PKIs) developed in oncology and to identify situations where these methods could be both feasible and useful. This ancillary study used data from a systematic review conducted to identify dose-finding studies for PKIs. PKIs selected for meta-analyses were required to have at least five completed dose-finding studies involving cancer patients, with available results, and dose escalation guided by toxicity assessment.

Background: Methodological limitations affect a significant number of oncology and haematology trials, raising concerns about the applicability of their results. For example, a suboptimal control arm or limited access to best care upon progression may skew the trial results toward a benefit in the experimental arm. Beyond the fact that such limitations do not prevent drugs reaching the market, other assessment tools, such as those developed by professional societies-ESMO-MCBS and ASCO Value Framework-do not integrate these important shortcomings.

Article Synopsis
  • Sepsis is a critical condition that can lead to severe illness and death, requiring prompt identification and treatment, but existing methods like the quick-SOFA score have limitations in effectiveness.* -
  • This study involved 796 patients suspected of community-acquired infections in the emergency department, from which a new clinical score was developed using various predictors to better identify sepsis risks.* -
  • The new score demonstrated a strong performance with an area under the ROC curve of 0.85, significantly outperforming existing scores like qSOFA, indicating it may be a more reliable tool for early sepsis detection.*
