In head and neck cancers, the effectiveness of cisplatin (CisPt) treatment is limited by its toxicity, especially when higher doses are necessary, and the possible occurrence of cisplatin resistance. This study evaluated the effects of resveratrol (RSV) on the expression of different genes involved in the response of human tumor cells (FaDu, PE/CA-PJ49) to cisplatin therapy. Our results revealed that RSV induced apoptosis amplification in both FaDu and PE/CA-PJ49 cells and modulated the expression of specific genes differently than in normal HaCaT cells.

Macrophages and Stem Cells-Two to Tango for Tissue Repair?

Biomolecules

May 2021

Cell Biology, Neurosciences and Experimental Myology Laboratory, Victor Babeș Institute of Pathology, 050096 Bucharest, Romania.

Macrophages (MCs) are present in all tissues, not only supporting homeostasis, but also playing an important role in organogenesis, post-injury regeneration, and diseases. They are a heterogeneous cell population due to their origin, tissue specificity, and polarization in response to aggression factors, depending on environmental cues. Thus, as pro-inflammatory M1 phagocytic MCs, they contribute to tissue damage and even fibrosis, but the anti-inflammatory M2 phenotype participates in repairing processes and wound healing through a molecular interplay with most cells in adult stem cell niches.

Article Synopsis
  • Treating superficial wounds is usually straightforward for healthy people, but more severe wounds like burns and diabetic ulcers present significant challenges for healthcare providers.
  • Our research focused on the effectiveness of creams and hydrogels in wound healing, highlighting that both can help prevent infections and promote recovery, but their success depends on the wound's characteristics and the patient's unique situation.
  • Hydrogels are particularly advantageous as they can incorporate various active ingredients and maintain their effectiveness through controlled release, and combining these with innovative therapies could enhance treatment outcomes when done correctly.

The immune system is dysfunctional in cancer, and therapeutic approaches designated to restore immunity and increase long-term overall survival are desirable. The role of immunotherapy is to trigger the immune system to recognize and destroy tumor cells. Interleukin-15 (IL-15) is a member of the common gamma-chain (γc) cytokines that promote the differentiation and expansion of T cells, B cells and natural killer (NK) cells, leading to enhanced antitumor responses.

Muscular dystrophy: Experimental animal models and therapeutic approaches (Review).

Exp Ther Med

June 2021

Laboratory of Cell Biology, Neuroscience and Experimental Myology, 'Victor Babes' National Institute of Pathology, 050096 Bucharest, Romania.

The muscular dystrophies are a heterogeneous group of genetically inherited diseases characterized by muscle weakness and progressive wasting, which can cause premature death in severe forms. Although >30 years have passed since the identification of the first protein involved in a type of muscular dystrophy, there is no effective treatment for these disabling disorders. In the last decade, several novel therapeutic approaches have been developed and investigated as promising therapeutic approaches aimed to ameliorate the dystrophic phenotype either by restoring dystrophin expression or by compensating for dystrophin deficiency.

An exosomal-carried short periostin isoform induces cardiomyocyte proliferation.

Theranostics

July 2021

Laboratory of Cellular and Molecular Cardiology, Istituto Cardiocentro Ticino, Lugano, Switzerland.

Although a small number of cardiomyocytes may reenter the cell cycle after injury, the adult mammalian heart is incapable of a robust cardiomyocyte proliferation. Periostin, a secreted extracellular matrix protein, has been implicated as a regulator of cardiomyocyte proliferation; however, this role remains controversial. Alternative splicing of the human periostin gene results in 6 isoforms lacking sequences between exons 17 and 21, in addition to full-length periostin.
