Exogenous genes are inserted into target cells during gene therapy in order to compensate or rectify disorders brought on by faulty or aberrant genes. However, gene therapy is still in its early stages because of its unsatisfactory therapeutic effects which are mainly due to low transfection efficiency of vectors, high toxicity, and poor target specificity. A natural polymer with numerous bioactive sites, good mechanical qualities, biodegradability, biocompatibility, and processability called silk fibroin has gained attention as a possible gene therapy vector.

Objective: To provide insights and strategies for pegvaliase management in challenging cases with phenylketonuria (PKU) based on the first 5 years of experience with pegvaliase in real-world clinical practice.

Methods: Twelve PKU experts gathered during a one-day, in-person meeting to discuss clinical cases illustrating important lessons from their experiences treating patients with pegvaliase in real-world clinical practice. Challenges with pegvaliase experienced prior to and during treatment and corresponding strategies to overcome them were discussed.

Topological interface states (TISs), known for their distinctive capabilities in manipulating electromagnetic waves, have attracted significant interest. However, in conventional all-dielectric one-dimensional photonic crystal (1DPC) heterostructures, TISs strongly depend on incident angle, which limits their practical applications. Here, we realize an angle-independent TIS in 1DPC heterostructures containing hyperbolic metamaterials (HMMs) for transverse magnetic polarized waves.

Parasites can significantly reduce host fitness, affecting reproduction, growth, and survivability. Vertebrate hosts exhibit defensive strategies against parasites, including "sickness behaviors" such as lethargy and self-grooming to remove ectoparasites.
