A new sulfated triterpene glycoside with the sulfate group located in an unusual position in the carbohydrate moiety, was isolated from the MeOH extract of the aerial parts of Bupleurum rigidum. This compound was identified by a combination of chemical degradation and spectral methods as 3beta,16beta,23-trihydroxy-13, 28-epoxyolean-11-en-3beta-yl-beta-D-glucopyranosyl-(1-->2)[4-sulfate- beta-D-glucopyranosyl-(1-->3)]-beta-D-fucopyranoside (sandrosaponin I) (1). In addition, the known compound 3beta,16beta, 23-trihydroxy-13, 28-epoxyolean-11-en-3beta-yl-beta-D-glucopyranosyl-(1-->2)[beta-D-glu copyranosyl-(1-->3)]beta-D-fucopyranoside (2) was isolated in the present investigation.

Background And Aims: The genus Bupleurum has long been recognized as a natural group, but its infrageneric classification is controversial and has not yet been studied in the light of sequence data.

Methods: Phylogenetic relationships among 32 species (35 taxa) of the genus Bupleurum were investigated by comparative sequencing of the ITS region of the 18-26S nuclear ribosomal DNA repeat. Exemplar taxa from all currently accepted sections and subsections of the genus were included, along with outgroups from four other early branching Apioideae genera (Anginon, Heteromorpha, Physospermum and Pleurospermum).

As part of our screening of antiviral agents from medicinal plants, 11 compounds from plant origin (Bupleurum rigidum and Scrophularia scorodonia), three saikosaponins, seven iridoids and one phenylpropanoid glycoside were tested in vitro against herpes simplex type I (HSV-1), vesicular stomatitis virus (VSV) and poliovirus type 1. Five of these compounds showed antiviral activity against VSV. The percentages of cellular viability at the non-toxic limit concentrations of the active compounds were: verbascoside 53.

Five triterpenoid saponins [buddlejasaponin IV, sandrosaponins VII (1), VIII (2), IX (3), and X (4)] were isolated from an n-BuOH extract of the roots of Bupleurum rigidum. Sandrosaponins VII-X (1-4) are new compounds, and their structures were established by 1D and 2D NMR techniques, FABMS, and chemical methods.
