Conformational switching in an aspartic proteinase.

The crystal structure of a catalytically inactive form of cathepsin D (CatDhi) has been obtained at pH 7.5. The N-terminal strand relocates by 30 A from its position in the interdomain beta-sheet and inserts into the active site cleft, effectively blocking substrate access. CatDhi has a five-stranded interdomain beta-sheet and resembles Intermediate 3, a hypothetical structure proposed to be transiently formed during proteolytic activation of the proenzyme precursor. Interconversion between active and inactive forms of CatD is reversible and may be regulated by an ionizable switch involving the carboxylate side chains of Glu 5, Glu 180, and Asp 187. Our findings provide a structural basis for the pH-dependent regulation of aspartic proteinase activity and suggest a novel mechanism for pH-dependent modulation of substrate specificity.

Previous studies have yielded inconsistent findings regarding the comparison of lipid profiles in patients with primary aldosteronism (PA) and essential hypertension (EH), and there is a limited understanding of the association between plasma aldosterone concentration (PAC) and lipid profiles across various populations. 270 patients with PA and 598 patients with EH diagnosed at the Second Affiliated Hospital of Nanchang University from January 2020 to June 2021 were consecutively included. In this work, we describe the synthesis of three new -arylporphyrins, named -tetrakis [4-(nicotinoyloxy)phenyl] porphyrin (), -tetrakis [4-(picolinoyloxy)phenyl] porphyrin (), and -tetrakis [4-(isonicotinoyloxy) phenyl] porphyrin (). These new synthesized -arylporphyrins are characterized using spectroscopic analysis: Fourier Transform Infrared Spectroscopy (FTIR) and One-dimensional Nuclear Magnetic Resonance (1D NMR), and mass spectrometry (MS). Signal peptide peptidase (SPP) is an ER-resident aspartyl intramembrane protease cleaving proteins within type II-oriented transmembrane segments. Here, we identified the tail-anchored protein Three prime repair exonuclease 1 (TREX1) as a novel substrate of SPP. Based on its DNase activity, TREX1 removes cytosolic DNA acting as a negative regulator of the DNA-sensing cGAS/STING pathway.
