Background: The relationship between gastric mucosal damage induced by stress, peptides present in the gastric mucosa and is not clear. Aim of this study was to determine whether cold-restraint stress affected the release of gastric somatostatin, gastrin and in the isolated perfused stomach preparation.

Methods: Male Sprague-Dawley rats were used, 12 cold-restraint stressed and 12 unstressed controls. 4 additional unstressed rats were treated with aspirin (100 mg/kg p.o.). After 30 minutes, isolated stomachs were perfused for 50 minutes with Krebs-Ringer buffer added with isoproterenol or carbamylcholine plus somatostatin-14 or carbamylcholine alone, somatostatin, gastrin and prostaglandin E2 release in the portal vein effluent were measured by radioimmuno-assay. Histology of the gastric mucosa was obtained from a further 4 stressed and 4 unstressed rats.

Results: In the stomach from stressed animals, the somatostatin response to isoproterenol and the prostaglandin E2 response to carbamylcholine plus somatostatin were significantly lower than in the controls, whereas gastrin response to carbamylcholine was enhanced by stress. Treatment with aspirin abolished the prostaglandin E2 response to stimulation. Gastric mucosa histology from stressed and unstressed animals showed no significant lesions.

Conclusions: The inhibition of gastric somatostatin and prostaglandins release coupled to an enhanced acid stimulatory influence appear to antidate gastric mucosal injury and should play a role in the stress ulcer genesis.

Article Synopsis
  • The article evaluates the effectiveness and safety of a combination treatment of atropine (ATR) and omeprazole (OME) for acute gastritis (AG) in comparison to anisodamine (ADM) with OME.
  • The study involved 95 patients, with the ATR+OME group showing a higher success rate, fewer side effects, and quicker symptom relief than the ADM+OME group.
  • Results indicated that the ATR+OME treatment significantly reduced inflammatory markers and gastrointestinal hormone levels, suggesting its strong efficacy and safety for managing AG, making it suitable for wider clinical use.

Protective Effect of Pentoxifylline on the Development of Acute Gastric Mucosal Injury in a Model of LPS-Induced Sepsis.

Antioxidants (Basel)

December 2024

Department of Biochemistry and Molecular Biology, School of Medicine, Complutense University of Madrid, Avda. Complutense, s/n, 28040 Madrid, Spain.

Alterations in the gastric mucosal barrier, one of whose fundamental components is phosphatidylcholine (PC), may play an important role in the pathophysiology of erosive gastritis secondary to sepsis. Pentoxifylline (PTX) has been shown to reduce tissue damage in various experimental models of sepsis. The aim of this study was to investigate the effect of PTX on gastric mucosa PC synthesis, leukocyte infiltration, arachidonic acid-related metabolites, inflammation, oxidative stress, NO, CO, and somatostatin in a rat model of LPS-induced sepsis.

Bone and muscle mass characteristics in patients with gastroenteropancreatic neuroendocrine neoplasms.

Endocrine

December 2024

Εndocrinology Unit, First Department of Propaedeutic and Internal Medicine, National and Kapodistrian University of Athens, Medical School, Athens, Greece.

Background: Neuroendocrine neoplasms (NEN) are rare tumors arising from neuroendocrine cells most commonly in the gastrointestinal-tract. In recent years, advancements in therapeutics have increased survival rates in patients with NEN leading to a greater clinical burden compared to the general population.

Methods: The aim of this single-center case-control study was to investigate the incidence of low bone mass and changes in body composition in adult patients diagnosed with gastroenteropancreatic neuroendocrine tumors (GEPNET).

Article Synopsis
  • The study aimed to assess how Weichang'an pill (WCAP) affects functional dyspepsia (FD) in rats, focusing on brain-gut peptides and gut microbiota as potential treatment mechanisms.
  • Researchers created an FD rat model and divided the rats into different treatment groups, including those receiving WCAP and a control for comparison, then measured gastrointestinal motility and gut microbiota composition.
  • Results showed that WCAP improved FD symptoms by enhancing gastric emptying, regulating brain-gut peptide levels, and restoring gut microbiota balance, suggesting it could be an effective treatment for FD.
