The cloning of melanocortin receptors opened new avenues to identify selective ligands for this receptor family. gamma-MSH was characterized as a melanocortin-3 receptor selective agonist, [D-Arg8]ACTH-(4-10) and [Pro8,10, Gly9]ACTH-(4-10) were characterized as melanocortin-4 receptor antagonists. The application of these ligands in vivo revealed that melanocortin-4 receptors mediate melanocortin-induced grooming behaviour in the rat. Since we still lack potent and selective melanocortin receptor ligands, we performed homology modelling and site directed mutagenesis of the melanocortin-4 receptor, in order to understand how melanocortins bind melanocortin receptors. A histidine at position 260 in the melanocortin-4 receptor is important for normal receptor function. However this residue is not forming a salt bridge with a glutamate at position 92 to keep the receptor in an inactive conformation, nor with the glutamate in the melanocortin peptides as had been suggested before.

Obesity is increasingly prevalent worldwide, with genetic factors contributing to its development. The hypothalamic leptin-melanocortin pathway is central to the regulation of appetite and weight; leptin activates the proopiomelanocortin neurons, leading to the production of melanocortin peptides; these in turn act on melanocortin 4 receptors (MC4R) which suppress appetite and increase energy expenditure. MC4R mutations are responsible for syndromic and non-syndromic obesity.

Fluoxetine is a selective serotonin reuptake inhibitor (SSRI) widely used for depression, but its potential effects on gut microbiota regulation and vagus nerve-mediated serotonin receptor expression have not been well studied. We investigated changes in the gut microbiome regulated by fluoxetine and vagus nerve-mediated expression of several serotonin (5-HT) receptor types associated with anxiety and depression. Oral administration of fluoxetine alleviated lipopolysaccharide (LPS)-induced depressive and anxiety behaviors, increased 5-HT1A, 2 C, and melanocortin 4 (MC4) receptor expression, and the composition of Lactobacillus in mice's gut microbiome.
