The sedimentation characteristics of polysomal mRNA labelled in vitro by [5-3H]uridine and electrophoretic mobility of similar non-labelled mRNA of mouse plasmacytoma were studied. Rapidly labelled polysomal mRNA may be considered as mRNA on the basis of several independent but indirect tests. These mRNA's are localized in 18-6S region of sucrose gradient. Some part of radioactivity have been found in the ribosomal RNA. It was shown that there is 8-10 RNA fractions in sucrose gradient. The 16S and 12-14S fractions are isolated and partially purified by two- and three-fold centrifugation. Fractions homogenous in sucrose gradient were electrophoresed in PAAG-SDS and divided into several subfractions some of them being common for 16S and 12-14S. The number of non-crossover subfractions was about 2-3. Not less than 20 different main fractions of polysomal mRNA were determined in plasmacytoma cells on the basis of electrophoretic data.

GEMIN5 is a predominantly cytoplasmic multifunctional protein, known to be involved in recognizing snRNAs through its WD40 repeats domain placed at the N-terminus. A dimerization domain in the middle region acts as a hub for protein-protein interaction, while a non-canonical RNA-binding site is placed towards the C-terminus. The singular organization of structural domains present in GEMIN5 enables this protein to perform multiple functions through its ability to interact with distinct partners, both RNAs and proteins.

Translational silence of spermatozoa has long been considered the norm in animals. However, studies in mammals have shown that the mitochondrial ribosomal machinery is selectively activated during capacitation in the female reproductive tract, while cytosolic ribosomes remain inactive. Here, using quantitative proteomics in a piscine model species, we show that proteins involved in mRNA processing and cytoplasmic translation are predominantly accumulated in immature spermatozoa within the extratesticular excurrent ducts, while those related to flagellar motility are enriched in ejaculated (mature) sperm.

Pseudouridine (Ψ) is the most abundant RNA modification, yet studies of Ψ have been hindered by a lack of robust methods to profile comprehensive Ψ maps. Here we utilize bisulfite-induced deletion sequencing to generate transcriptome-wide Ψ maps at single-base resolution across various plant species. Integrating ribosomal RNA, transfer RNA and messenger RNA Ψ stoichiometry with mRNA abundance and polysome profiling data, we uncover a multilayered regulation of translation efficiency through Ψ modifications.
