A potential two-way passage of cells and substances between the brain and skull bone marrow may open for new insights into neurological disease. The arachnoid membrane was traditionally considered to restrict cells and larger molecules in CSF from entering the dura and bone marrow directly. However, new data on exchange between brain and skull bone marrow have recently emerged.

The two obstacles for treating glioma are the skull and the blood brain-barrier (BBB), the first of which forms a physical shield that increases the difficulties of traditional surgery or radiotherapy, while the latter prevents antitumor drugs reaching tumor sites. To conquer these issues, we take advantage of the high penetrating ability of sonodynamic therapy (SDT), combined with a novel nanocomplex that can easily pass the BBB. Through ultrasonic polymerization, the amphiphilic peptides (CGRRGDS) were self-assembled as a spherical shell encapsulating a sonosensitizer Rose Bengal (RB) and a plant-derived compound, sulforaphane (SFN), to form the nanocomplex SFN@RB@SPM.

Skulls with high optical scattering and acoustic attenuation are a great challenge for photoacoustic imaging for human beings. To explore and improve photoacoustic generation and propagation, we conducted the photoacoustic simulation and image reconstruction of the multi-layer brain model with an embedded blood vessel under different optical source types. Based on the optical simulation results under different types of optical sources, we explored the characteristics of reconstructed images obtained from acoustic simulations with and without skull conditions.
