Synthesis and pharmacological evaluation of ring-methylated derivatives of 3,4-(methylenedioxy)amphetamine (MDA).

The three isomeric ring-methylated derivatives of the well-known hallucinogen and entactogen MDA (1a) were synthesized and evaluated for pharmacological activity as monoamine-releasing agents and as serotonin agonists. The 2-methyl derivative 2a and the 5-methyl derivative 2b were found to be more potent and more selective than the parent compound in inhibiting [3H]-serotonin accumulation in rat brain synaptosomal preparations. Their activity in vivo was confirmed in rats trained to discriminate serotonin-releasing agents and hallucinogens from saline. The results indicate that compounds 2a,b are among the most potent 5-HT-releasing compounds known and show promise as lead compounds in the search for antidepressant drugs that release serotonin rather than inhibit its uptake.

Psilocin (1) is the dephosphorylated and psychotropic metabolite of the mushroom natural product psilocybin. Oxidation of the phenolic hydroxy group at the C-4 position of 1 results in formation of oligomeric indoloquinoid chromophores responsible for the iconic blueing of bruised psilocybin-producing mushrooms. Based on previous NMR experiments, the hypothesis included that the 5,5'-coupled quinone dimer of 1 was the primary product responsible for the blue color.

Alkylresorcinols (5-alkyl-1,3-dihydroxybenzenes, ARs) are bioactive phenolic lipid compounds which are particularly abundant in rye and partly other cereals. In this study on ARs, whole rye grain extracts were gained with cyclohexane/ethyl acetate (46/54, w/w). Silylated extracts were used to develop a gas chromatography with mass spectrometry method in the selected ion monitoring mode (GC/MS-SIM) for the sensitive detection of conventional ARs along with keto-substituted (oxo-AR) and ring-methylated ARs (mAR) with 5-alkyl chain lengths of 14 to 27 carbon atoms and 0 to 4 double bonds in one run.

It has already been reported that in vivo muscle necrosis induced by various phenylenediamine derivatives correlated with their in vitro autoxidation rate [9]. Now in a more detailed investigation of the cytotoxic mechanism of a ring-methylated phenylenediamine known as tetramethylphenylenediamine or durenediamine (DD) towards isolated rat hepatocytes has been carried out. Cytotoxicity was preceded by ROS formation which was markedly increased by inactivating DT-diaphorase or catalase but were prevented by a subtoxic concentration of the mitochondrial respiratory inhibitor cyanide.

A series of novel carbocations were generated from isomeric monoalkylated and dialkylated benz[a]anthracenes (BAs) by low-temperature protonation in FSO(3)H/SO(2)ClF. With the monoalkyl derivatives (5-methyl, 6-methyl, 7-methyl, and 7-ethyl) as well as the D-ring methylated analogues (9-methyl, 10-methyl, and 11-methyl), the C-7 or the C-12 protonated carbocations were observed (as the sole or major carbocation) in all cases. Protonation of the 12-methyl derivative (9) gave the C-7 protonated carbocation (9H+) as the kinetic species and the ipso-protonated carbocation (9aH+) as the thermodynamic cation.

The ground state conformation, spectroscopy, and photochemical behavior of styrene and eight of its vinyl- and ring-methylated derivatives have been investigated. Introduction of methyl groups at the alpha-position of the vinyl group or the ortho positions of the phenyl results in increased phenyl-vinyl dihedral angles. Styrenes possessing both alpha-methyl and ortho-methyl groups adopt orthogonal geometries.
