Cryopreserved erythrocytes with the period of storage from 5 to 12 months were studied in 10 cardiosurgical patients under conditions of scanning electron microscopy. It was established that among unfrozen erythrocytes of patients with rheumatic diseases of the heart there were (84.25 +/- 0.7)% of normocytes and only (4.25 +/- 1.03)% of deformed cells. During the experimental perfusion the amount of normocytes decreased to (57.50 +/- 1.80)% and the amount of destroyed normocytes inconsiderably increased by the 4th hour of perfusion. The intermediate forms of erythrocytes had a clear tendency to an increase of their amount. The results obtained can show that cryoerythrocytes of cardiosurgical patients are sufficiently resistant to unfavorable factors of the artificial blood circulation, are not inferior to donor erythrocytes by their morphological properties and can be taken as a basis for transfusion therapy in operations on the open heart.

Background: We investigated the intraneural spread of injected fluid in brachial plexus nerve roots, examining the potential for intrafascicular spread and identifying influencing factors.
Methods: Twelve deliberate ultrasound-guided intraneural injections were performed at the ventral rami of the brachial plexus nerve roots at their exits from the neuroforamina in six fresh, unembalmed, cryopreserved human cadavers. A 22-G, 30-degree bevel echogenic regional anaesthesia needle was used.

Background: The ACP 215 automated cell processor is used to glycerolize and deglycerolize red cell concentrates (RCCs). Its primary advantage over the COBE 2991, previously used to cryopreserve RCCs, is that it maintains a closed system enabling extended post-thaw expiry. However, it was observed that post-deglycerolization hematocrits (Hct) of units processed with the LN236 kit are markedly lower than those processed using the COBE 2991.

Initially observed on synthetic nanoparticles, the existence of biomolecular corona and its role in determining nanoparticle identity and function are now beginning to be acknowledged in biogenic nanoparticles, particularly in extracellular vesicles - membrane-enclosed nanoparticle shuttling proteins, nucleic acids, and metabolites which are released by cells for physiological and pathological communication - we developed a methodology based on fluorescence correlation spectroscopy to track biomolecular corona formation on extracellular vesicles derived from human red blood cells and amniotic membrane mesenchymal stromal cells when these vesicles are dispersed in human plasma. The methodology allows for tracking corona dynamics under physiological conditions. Results evidence that the two extracellular vesicle populations feature distinct corona dynamics.

Controlling the formation and growth of ice is essential to successfully cryopreserve cells, tissues and biologics. Current efforts to identify materials capable of modulating ice growth are guided by iterative changes and human intuition, with a major focus on proteins and polymers. With limited data, the discovery pipeline is constrained by a poor understanding of the mechanisms and the underlying structure-activity relationships.
