Storage of single-donor platelets is currently limited to 5 days. During this period, however, numerous morphologic and biochemical changes have been observed. The present study describes increased binding of anti-GMP 140 to stored single-donor platelet concentrates; this reveals a progressive activation process. In contrast, when stored platelets are stimulated with ADP, GMP 140, expression is reduced indicating a diminished release reaction. Additionally, the GP IIb/IIIa receptor complex and its fibrinogen-induced binding site (LIBS1) are reduced by storage time, demonstrating a diminished fibrinogen binding. Further experiments have to clarify whether these changes observed in vitro translate into a reduced hemostatic capacity after transfusion in vivo.

In the process of preparation and storage of platelets, centrifugation, leukofiltration, and agitation will cause morphological changes and impaired function of platelets, which is associated with the increase of platelet transfusion refractoriness, and named as platelet storage lesion (PSL).

Method: This paper proposes three major operations (centrifugation, agitation, and leukofiltration) that platelets experience during the preparation and storage process, to explore the effect of physical cues on PSL.

Using platelet concentrates to treat maxillofacial tissue lesions.

Objectives: Platelet concentrates (PCs), which are blood products that are abundant in platelets and growth factors, have become pivotal in treating maxillofacial tissue lesions due to their capacity for promoting bone and soft tissue recovery. This review will provide some recent progress of the use of platelet concentrates to treat lesions on maxillofacial tissues.

Subjects: We reviewed the mechanisms by which PCs promote wound healing and tissue recovery and summarized the application of PCs in the treatment of lesions on maxillofacial tissues, including medication-related osteonecrosis of the jaw, post-extraction wound healing, implant surgery, temporomandibular joint diseases, and periodontal tissue restoration.
