The use of cryopreserved donor spermatozoa for insemination has become necessary to decrease the risk from sexually transmitted infectious diseases. Lower fecundity rates have been reported with this practice. Efforts have been applied to increase success, including identification of those sperm characteristics which correlate with increased fecundity. Data from in-vitro fertilization have revealed sperm morphology, motility and zona pellucida binding as important sperm parameters. Discontinuous Percoll gradient preparation yields a high concentration of motile spermatozoa. Using this preparation for thawed donor spermatozoa, we have identified post-preparation motility and progression as factors associated with increased fecundity. Consideration should be given to screening sperm donors with a freeze-thaw Percoll gradient preparation prior to acceptance into a donor bank.

This study evaluated the effectiveness of Percoll® density gradient centrifugation (Percoll-DGC) for selecting bull epididymal sperm prior to conventional slow (CS) or ultra-rapid (UR) freezing and its effects on sperm quality. Fifteen pooled samples from 30 epididymides (2 different samples/pool) of 15 bulls were split into two aliquots assigned to either CS or UR freezing. The design of chemomechanical self-oscillators, which execute oscillations in the presence of constant stimuli lacking periodicity, is a step toward the development of autonomous and interactive soft robotic systems. This work presents a simple design of prolonged chemomechanical oscillatory movement in a microgel system capable of buoyant motility within stratified chemical media containing spatially localized sinking and floating stimuli. Three design elements are developed: a stimuli-responsive membranized calcium alginate microgel, a Percoll density gradient for providing stratified antagonistic chemical media, and transduction of microgel particle size actuation into buoyant motility via membrane-mediated displacement of the Percoll media.
