As part of an effort to prepare efficacious and orally bioavailable analogs of the previously reported thrombin inhibitors 1a, b, we have synthesized a series of compounds that utilize 3,3-disubstituted propionic acid derivatives as P3 ligands. By removing the N-terminal amino group, the general oral bioavailability of this class of compounds was enhanced without excessively increasing the lipophilicity of the compounds. The overall properties of the molecules could be drastically altered depending on the nature of the groups substituted onto the 3-position of the P3 propionic acid moiety. A number of the compounds exhibited good oral bioavailability in rats and dogs, and numerous compounds were efficacious in a rat FeCl3-induced model of arterial thrombosis. Compound 7, the 3,3-diphenylpropionic acid derivative, showed the best overall profile of in vivo and in vitro activity. Molecular modeling studies suggest that these compounds bind in the thrombin active site in a manner essentially identical to that previously reported for compound 1a.

Probiotics and their postbiotics have the potential to improve the health and growth performance of piglets, which has brought them widespread attention in the post-antibiotic era. In the present study, the effects of dietary supplementation of postbiotics on the growth performance, intestinal flora structure and plasma metabolome of weaned piglets were investigated. Attention-deficit/hyperactivity disorder (ADHD), a common neurodevelopmental disorder in children, is associated with alterations in gut microbiota and short-chain fatty acids (SCFAs), which are metabolites influencing the gut-brain axis. Evidence suggests that psychostimulant medications, widely used to manage ADHD symptoms, may also impact gut microbiota composition and SCFA levels. This study explores these potential effects by examining gut microbiota profiles and SCFA concentrations in unmedicated and medicated children with ADHD, compared to healthy controls.

The current study investigated the in vitro degradability, in vitro gas production, methane (CH) production, and ruminal bacterial community of kenaf plants cut at different heights (130, 160, 190, 220, and 250 cm). These samples were subjected to an in vitro batch culture system using buffalo rumen fluid to measure gas and CH production at 3, 6, 9, 12, 24, 36, 48, and 72 h of incubation. Results reveal that crude protein (CP) concentration was the highest at the 220 cm height compared with the other heights.
