In 8 patients with idiopathic chronic urticaria suction blisters were produced simultaneously on lesional and perilesional skin in the periumbilical area without application of heat. The suction pressure was 250 mm Hg, and thus the blister production lasted from 60--120 min. The blister fluid histamine was determined with a modified autoanalyzer technique developed by Siraganian. In this method the extractionof histamine, the condensation of o-phthaldial-dehyde with histamine and the measurement of the fluorochrome is automated. With the described method it is possible to measure values less than 0.5 ng/ml. Histamine levels in perilesional skin were nearly in the same range as in the periumbilical skin of 10 normal persons. The range of the normal tissue fluid histamine levels was 0--15 ng/ml and they did not differ significantly from the plasma levels. In all 8 patients with chronic urticaria the histamine values were higher in lesional than in perilesional skin. In 1 patient the lesional blister fluid histamine value reached 35.5 ng/ml. Four of the patients showed only low differences (less than 3 ng/ml).

Expression of JAK/STAT signalling in bullous pemphigoid.

Australas J Dermatol

November 2024

Department of Dermatology, Eastern Health, Monash University, Melbourne, Victoria, Australia.

In the present study, we characterise the expression of JAK/STAT signalling in Bullous Pemphigoid (BP). Punch biopsy specimens of normal skin, lesional and perilesional skin of patients diagnosed with biopsy proven BP were utilised for immunoperoxidase labelling with commercially available antibodies for Phosphorylated STAT 1,2,3,5A and 6 (Genesearch, USA). Quantitative analysis demonstrated that in comparison to normal skin, phosphorylated STAT 2, 3, 5A and 6 were overexpressed in BP.

Skin microbiota plays an essential role in the development and function of the cutaneous immune system, in the maintenance of the skin barrier through the release of antimicrobial peptides, and in the metabolism of some natural products. With the aim of characterizing changes in the cutaneous microbiota specifically associated with wound healing in the diabetic condition, we performed a 16 S rRNA gene Next Generation Sequencing of skin swabs taken within the ulcer bed of ten diabetic patients before (t0) and after 20 days of therapy (t20) with a fluorescein-based galenic treatment. Considering the twenty most representative genera, we found at t20 an increase of Corynebacterium, Peptostreptococcus, and Streptococcus, and a decrease of Enterococcus, Finegoldia, and Peptoniphilus genera.

Introduction: Melanoma, a malignant tumor derived from neural crest melanocytes, predominantly affects the skin but can involve any organ with neural crest migration. Metastatic melanoma of unknown origin, particularly when it involves the brain, is associated with significant morbidity, mortality, and a typically poor prognosis.

Case Presentation: The authors present a 71-year-old man with a history of hypertension and seizure disorder who experienced a headache, transient loss of consciousness, and vomiting.

Article Synopsis
  • The study investigates the role of oxidative stress and IFNγ-driven immune responses in the development of vitiligo, focusing on how mast cells produce IFNγ and its effects on the disease progression.* -
  • Researchers analyzed skin samples from vitiligo patients and used a mouse model to explore how stress signals (like hydrogen peroxide) affect the recruitment of mast cells and CD8+ T cells to affected skin areas.* -
  • Results showed that IFNγ-producing mast cells increased alongside CD8+ T cells in early vitiligo, suggesting that skin stress leads to changes in immune cell dynamics, which may contribute to the pathogenesis of vitiligo.*
