Huntington's disease (HD) arises from the abnormal expansion of a CAG repeat in the HTT gene. The mutant CAG repeat triggers aberrant RNA-protein interactions and translates into toxic aggregate-prone polyglutamine protein. These aberrant RNA-protein ineractions also seed the formation of cytoplasmic liquid-like granules, such as stress granules.

Methods: Our institution's ACL Registry was consulted to identify patients between the age of 18 and 45 who underwent primary ACL reconstruction between Jan 2019 and July 2022 for acute, noncontact ACL rupture. Patients with pre-existing meniscal pathology, chronic ACL reconstructions, revisions, and multi-ligament knee injuries were excluded.

Automating the process of spotting them in micrographs can significantly enhance research and monitoring. An analytical platform was developed using agarose gel to overcome the issues of device miniaturization and interference effects caused by foreign proteins, which are challenges for field analysis-oriented measurement technology. The lab-on-gel (LoG) device integrates sample preparation, reaction, and detection on a chip, with an agarose gel for separation that preferentially allows only small molecules to pass through due to the size exclusion effect, and an agarose gel for sensing that contains glucose dehydrogenase and substrates. Measurements can be performed simply by dropping in a diluted serum sample and then placing the device in a custom-made compact Raspberry Pi-based fluorescence imager.
