The structures of triacylglycerols from sclerotia of the rye ergot Claviceps purpurea (Fries) Tul.

The triacylglycerol fraction of lipids extracted from sclerotia of the rye ergot Claviceps purpurea strain VKM F-2642D has been shown to comprise about 60% of the total lipids and to consist of at least seven constituents, namely, [R]-[R]-[R] (1.6% of the total fraction), [R-Ric]-[R]-[R] (27%), [R-Ric]-[R-Ric]-[R] (65%), [R-Ric]-[R-Ric]-[R-Ric] (4.8%), [R-Ric-Ric]-[R-Ric]-[R-Ric] (1.1%), [R-Ric-Ric]-[R-Ric-Ric]-[R-Ric] (0.3%), and [R-Ric-Ric-Ric]-[R-Ric-Ric]-[R-Ric-Ric] (0.2%) (acyl residues at the 1(3), 2 and 3(1) positions of the glycerol moiety are indicated; R = nonhydroxylated fatty acid (C16, C18) residue, Ric = ricinoleyl residue). Non-hydroxylated fatty acyls (R) are n-16:0 (25-50% of the total), 16:1 omega 7 (10-15%), n-18:0 (2-5%), 18:1 omega 9 (20-30%), and 18:2 omega 6,9 (10-35%). The second and the third of the listed triacylglycerols are composed dominantly (by 86% or more) of 1,2-di-[R]-3-[R-Ric]-sn-glycerol and 1-[R]-2,3-di-[R-Ric]-sn-glycerol, respectively.

Ergot alkaloids (EAs), mycotoxins produced mainly by fungi of the genus, have been frequently reported in rye, while their increasingly frequent occurrence in other cereals is likely related to weather conditions, with the incidence of ergot sclerotia in winter grains being related to heavy rainfall and moist soils at critical periods. However, compared to other regulated mycotoxins, data about the prevalence and occurrence of EAs in major and minor cereals harvested in the Mediterranean growing areas are still scant. In this regard, the current study reported the occurrence of EAs in 18 genotypes of winter cereals harvested over 3 years from an experimental field located in North Italy which were analyzed by HPLC-MS/MS.

Rye is a secondary crop that is characterized by a higher tolerance to climatically less favorable conditions than other cereal species. For this reason, rye was historically used as a fundamental raw material for bread production and as a supply of straw in northern parts of Europe as well as in mountain environments, such as Alpine valleys, where locally adapted landraces have continued to be cultivated over the years. In this study, rye landraces collected in different valleys in the Northwest Italian Alps have been selected as the most genetically isolated within their geographical contexts and cultivated in two different marginal Alpine environments.

In Mesopotamian wheat fields rye grew as weed and after certain developments rye reached Europe and there became a cultivable plant and the dominant grain in certain regions. With the rye also its parasite, the Claviceps purpurea, reached Europe. This ascomycete infects particular rye grains and in its developmental cycle grows to sclerotia bigger than each grain.

The plant parasitic fungus sensu lato produces sclerotia containing toxic ergot alkaloids and uncharacterized indole diterpenoids in grasses including cereals. The aim of this study was to detect as many peptide ergot alkaloids and indole diterpenoids in ergot sclerotia as possible by using a liquid chromatography-high-resolution mass spectrometry (LC-HRMS/MS) approach and applying filtering of diagnostic fragment ions for data extraction.
