The alpha 1-agonist phenylephrine in hepatocytes from mature and senescent rats, and the beta-agonist isoproterenol in hepatocytes from senescent rats, elicited a time-dependent, homologous desensitization of alpha 1- and beta-receptor-mediated glycogenolysis respectively, which was maximal after 20 min of exposure to the agonists. Glucagon triggered desensitization of the glycogenolytic response to glucagon, isoproterenol and phenylephrine, which was maximal after less than 5 min; this was followed by resensitization of the glucagon response only. After 20 min of treatment with phenylephrine or isoproterenol, no change in cellular distribution of alpha 1- or beta-receptors was noticed, using sucrose gradient centrifugation. After a 1-h exposure to both agonists, a shift of both receptors to a light density fraction was found, reflecting receptor internalization. Neither the rate of functional desensitization, nor the degree of receptor internalization was altered upon ageing. We conclude that functional desensitization and internalization of adrenergic receptors in rat hepatocytes are separate events in time and are largely unaffected by the ageing process. Thus, despite the absence of a beta-receptor-mediated glycogenolytic response in hepatocytes from mature rats, isoproterenol triggers the internalization of beta-receptors.

Objective: To evaluate the retention rate and other clinical criteria of four different restorative techniques for non-carious cervical lesions (NCCLs) after 4 years.
Methods: This is a prospective, randomized, double-blind, and split-mouth study evaluating four different adhesion strategies in non-carious cervical lesion restorations: adhesive restorative system (Scotchbond Universal Adhesive/Filtek Z350XT) without (SBU) and with selective enamel acid-etching (E-SBU), resin-modified glass-ionomer cements (Vitremer; RMGIC), and ethylenediaminetetraacetic as acid pretreatment (E-RMGIC). In total, 200 restorations were placed in 50 patients.

The Drosophila neuromuscular junction (NMJ) is a powerful genetic system that has revealed numerous conserved mechanisms for synapse development and homeostasis. The fly NMJ uses glutamate as the excitatory neurotransmitter and relies on kainate-type glutamate receptors and their auxiliary protein Neto for synapse assembly and function. However, despite decades of study, the reconstitution of NMJ glutamate receptors using heterologous systems has been achieved only recently, and there are no reports on the gating properties for the recombinant receptors.

Objective: To evaluate a prior application of an experimental desensitizer containing a hydroxyapatite-capsaicin composite (HAp-CAP) in different concentrations on hydrogen peroxide (HP) diffusion into the pulp chamber, bleaching efficacy (BE) and on physical and chemical alterations of the enamel surface after in-office bleaching.
Materials And Methods: Forty sound premolars were randomized in five groups according to each experimental condition (n = 8): no bleaching (negative control), only bleaching (positive control), 0.1%, 1% and 5% of HAp-CAP.
