An efficient modified adeno system to express foreign genes to the central nervous system was developed recently. This modified recombinant adenoviral vectors can be used successfully to deliver lacZ to the hypothalamic suprachiasmatic nucleus (SCN) which is composed of mammalian clock pacemaker. The expression of lacZ in the primary culture of SCN was dose-dependent and higher enough in nearly 100% of these cells. We also showed that viral toxicity and lacZ overexpression had no serious effects on the rhythmic expression of arginine vasopressin (AVP) release from the SCN cell culture.

Background: A critical need to increase Alzheimer's disease (AD) screening exists given rising incidence, new disease-modifying treatments, and ill-equipped primary care settings. This study assessed the feasibility of a novel, community-based AD screening program to increase cognitive and retinal-based assessments.
Methods: An observational study, supported by the Davos Alzheimer's Collaborative, assessed the utility of leveraging community-based settings to increase rates of cognitive assessment (conducted by Alzheimer Society (AS) social workers [SWs]) and RetiSpec's AI-based eye test in optometry settings to detect biologic signatures of AD (plus participant survey).
Introduction: Evaluation of functional dependence in activities of daily living (ADLs) and instrumental activities of daily living (iADLs) is necessary for dementia diagnosis. ADL and iADL questionnaires are typically employed, but with progressive cognitive impairment, a care partner must step in to assist with these tests, causing logistical burdens. Pre-screening tools that triage patients in need of formal functional assessment would optimize clinical workflows.

Background: New immunotherapies for early-stage Alzheimer's disease (AD) have ushered in fresh hope for AD research and clinical care, but also highlight barriers to AD screening and timely diagnosis in the US. Digital cognitive assessments could potentially streamline screening and referrals for AD treatment and/or clinical trials. We report preliminary data on the feasibility and acceptability of three digital cognitive approaches for older adults completing Annual Wellness or routine follow-up visits with a primary care provider (PCP).

The Life and Health Questionnaire (LHQ) is a 32-item questionnaire - written at a 6th-grade reading level - that captures information related to potentially modifiable lifestyle and psychosocial risk factors, and protective factors for cognitive decline and dementia. The objective of this study was to gain insights into the feasibility of the LHQ in primary care settings by evaluating the influence of cognition, age, and education on the LHQ completion times.
