The enzyme steroid 5 alpha-reductase (5 alpha R) catalyzes the reduction of testosterone (T) to 5 alpha-dihydrotestosterone (DHT). In this study, the baculovirus expression system was used to overexpress rat 5 alpha R type I isozyme (r5 alpha R 1). The full length of r5 alpha R1 cDNA was inserted into the Autographa californica nuclear polyhedrosis virus (Ac-MNPV) genome and expressed in Spodoptera frugiperda, Sf 21, insect cells. The expressed recombinant r5 alpha-R1 showed maximal enzymatic activity when the infected cells were harvested on day 3 of post-transfection. The K(m) values for NADPH and T were 17 microM and 2.7 microM, respectively. Inhibition of the recombinant r5 alpha R1 by N,N diethyl-4-aza-4-methyl-3-oxo-5 alpha-androstane-17 beta-carboxamide (4MA) was competitive with respect to the substrate (T), and a Ki of 3 nM was obtained. The enzyme was located primarily in the nuclear fraction, and the maximum velocity for the recombinant r5 alpha R1 in this fraction was 60 nmoles DHT/min/mg. Immunoblot analysis indicated a single immunoreactive band at 26 kDa, which corresponds to the molecular weight of r5 alpha R1. Photoaffinity labeling by [2'-32P]-2-azido-NAD P+ ([2'-32P]2N3-NAD P+) and [1,2(3)H] N-(benzylbenzoyl)-3-oxo-4-aza-4-methyl-5 alpha androstane-17 beta-carboxamide ([3H]-4MABP) also showed a labeled protein band at 26 kDa.

Dissecting cellulitis (DC) of the scalp is a chronic inflammatory condition marked by neutrophilic cicatricial alopecia, often linked to staphylococcal antigens. This case report details a 34-year-old male with scarring acne who developed DC following follicular unit extraction (FUE) approximately four months prior. Trichoscopic examination revealed brown pigmented dots, erythema, and melicerous crusts.

Steroidal 5α-reductase type 2 (S5αR2) is a key enzyme involved in the conversion of testosterone (TST) to dihydrotestosterone (DHT), a crucial process in the development of benign prostatic hyperplasia (BPH). Phytosterols (PSs), natural plant-derived compounds, have been proposed as potential inhibitors of S5αR2, but studies on their efficacy are limited. This study evaluates the inhibitory effects of three PSs (β-sitosterol, stigmasterol, and campesterol) on S5αR2 activity using a combined in vitro and in silico approach.

Article Synopsis
  • - This study presents a quick and precise technique for screening steroid 5-alpha reductase (S5αR) inhibitors in the plant Impatiens balsamina, which has historical uses in treating hair loss and skin issues.
  • - Utilizing a combination of S5αR inhibitory assays and LC-QTOF-ESI-MS/MS, the researchers successfully identified eight bioactive compounds, with the most effective ones being 2-methoxy-1,4-naphthoquinone and impateinol, which showed significantly better S5αR inhibitory effects compared to the drug finasteride.
  • - The method developed is not only efficient for rapid screening of herbal compounds but also assists in the safe isolation of bioactive markers

Article Synopsis
  • Androgenetic Alopecia (AGA) affects a significant number of men, with 30% experiencing it by age 30 and 50% by age 50.
  • A study compared the effectiveness of a combination treatment of topical finasteride 0.25% and minoxidil 5% versus minoxidil 5% alone, involving 164 male participants aged 30-60 with AGA.
  • Results showed that 86.7% of the combination treatment group experienced hair regrowth, compared to 69.1% in the minoxidil-only group, indicating the combination therapy is more effective for treating AGA.
