[Immunotherapy of solid tumors. Clinical studies].

In this study we analyze the major clinical trials of immunotherapy for solid tumors. Much progress have been made in reducing the side effects and the percentage of patients which respond has increased. In immunotherapy with lymphokines the innovative orientation consist in the administration of low doses or decreasing doses and by alternative ways as regards infusion and systemically. The use of immunotherapy to stimulate the specific immune response seems to represent the most promising field from a therapeutic point of view. Studies in the field of in vitro expansion of immunocompetent cells have obtained results in the simplification of the technique and in an increase of its efficiency; moreover, at the moment, many clinical trials are involving specific immunotherapy using autologous neoplastic cells altered with adjuvant substance and the results are promising with very few side effects. In the near future immunotherapy with specific tumor antigens is sure it will play a major role.

As an important member of the Siglec family, SIGLEC-15 plays an important role in osteoclast differentiation, bone remodeling, and tumor immune evasion. In the tumor microenvironment, SIGLEC-15 functions independently of the B7-H1/PD-1 pathway. In this study, the SIGLEC-15 fusion protein (SIGLEC-15-Fc) was successfully expressed and purified using a eukaryotic expression system.

Background: Prolgolimab is an IgG1 anti-PD-1 monoclonal antibody with the Fc-silencing 'LALA' mutation. The phase III DOMAJOR study assessed efficacy and safety of prolgolimab in combination with pemetrexed and platinum-based chemotherapy vs placebo in combination with pemetrexed and platinum-based chemotherapy as first-line treatment for advanced non-small cell lung cancer (NSCLC).

Methods: 292 patients with advanced non-squamous NSCLC were randomized 1:1 to receive 4 cycles of pemetrexed, platinum-based drug and either prolgolimab (3 mg/kg Q3W) or placebo followed by prolgolimab/placebo with pemetrexed until disease progression or toxicity (≤36 months).

Shallow whole-genome sequencing (sWGS) offers a cost-effective approach to detect copy number alterations (CNAs). However, there remains a gap for a standardized workflow specifically designed for sWGS analysis. To address this need, in this work we present SAMURAI, a bioinformatics pipeline specifically designed for analyzing CNAs from sWGS data in a standardized and reproducible manner.
