In this report we show that the mouse interferon (IFN)-gamma R1 and IFN-gamma R2 subunits expressed in hamster cells are capable of rendering the cells sensitive to mouse IFN-gamma as measured by induction of class I MHC antigens and the activation of the transcription factor Stat1 alpha. However, these cells showed no antiviral protection in response to IFN-gamma when challenged with vesicular stomatitis virus (VSV) but limited protection when challenged with encephalomyocarditis virus (EMCV). Furthermore, the cytoplasmic domains of the IFN-gamma R2 subunits, like the cytoplasmic domains of the IFN-gamma R1 chains, can be interchanged between species with no loss of biologic activity, demonstrating that the species-specific interaction of the IFN-gamma R1 and IFN-gamma R2 chains involves only the extracellular domains of the two proteins.

We previously showed that selected CSF biomarkers of neuronal injury (NfL, VILIP-1), AD neuropathology (p-tau181), and neuroinflammation (GFAP, MCP-1, sTREM2) measured at presentation associated with etiologic diagnoses and reliably differentiated patients with treatment-responsive causes of rapid progressive dementia. However, no differences were identified between CSF biomarkers in patients with rapid and typical progressive forms of AD/ADRD, leaving key questions unanswered concerning the mechanisms that drive rpAD/ADRD.

Background: Peripheral blood mononuclear cells (PBMCs) were obtained from patients across different stages of Parkinson's disease (PD) progression and stimulated ex vivo to develop biomarkers for predicting PD progression.
Method: PBMCs obtained at one time-point from patients with moderate stage PD (>5 years after diagnosis) (n = 30), early stage PD (<5 years after diagnosis) (n = 27), prodromal PD (rapid-eye-movement sleep behavior disorder patients) (n = 14), and healthy controls (HCs) (n = 9) were isolated from whole blood and cryopreserved. Samples were thawed, then pan-monocytes and T-cell populations were isolated from PBMCs and subjected to treatment with vehicle or IFN-γ.
