This study explores evolved Hyaluronidase, Lipase, and Elastase's identification, characterization, and therapeutic potential to enhance tissue regeneration and drug delivery systems in otolaryngology. Hyaluronidase variant H5 exhibited a turnover number (k_cat) of 1500 min, a 200 % increase over wild-type (500 min), demonstrating superior hyaluronic acid degradation. Similarly, lipase variant L2 reached 1200 min (400 min wild-type), and elastase variant E3 showed a turnover of 2200 min (1000 min wild-type).

Lung cancer (LC) is the predominant cause of cancer-related fatalities globally, with the highest death rates in both genders, primarily attributed to smoking. The non-kinase transmembrane cell surface glycoprotein, CD44, enhances LC cell migration and invasion, leading to drug resistance and an unfavorable prognosis. This research formulated a cisplatin-loaded gold nanoshell (HCP@GNS) integrated with hyaluronan (HCP@GNS@HA) to enhance targeting capability and realize a synergistic effect of chemo-photothermal therapy (chemo-PTT) against LC.

However, how ECM proteins and proteoglycans collectively regulate cellular processes has not been adequately explored. Here, we address this question by studying cytoskeletal and focal adhesion organization and dynamics on cells cultured on polyacrylamide hydrogels functionalized with Col, HA and a combination of Col and HA (Col/HA).
