Role of calcium-activated neutral protease (calpain) with diet and exercise.

Although the proteolytic events accompanying acute and chronic perturbations in striated muscle protein turnover remain to be fully elucidated, the purpose of this paper is to (a) review the chemistry of the nonlysosomal calpain-calpastatin system, and (b) provide evidence for the involvement of a nonlysosomal, calcium-activated neutral protease (calpain) in the response of skeletal muscle protein breakdown to altered nutritional status (diet composition; energy restriction) and increased periods of contractile activity (exercise). In reviewing the literature, it is apparent that calpain is involved in the protein catabolism which accompanies alterations in diet composition and/or energy restriction. The precise mechanism of calpain action remains to be elucidated; however, the role of altered metabolic status contributing to calcium imbalances is discussed relative to increasing protein degradation. Hypotheses for further investigation are provided in regard to identifying the targeting of selected proteins (and organelles) for degradation by calpain.

Introduction: Schistosomiasis is a neglected tropical disease that puts over 200 million people at risk, and prevention options are sparse with no approved vaccine. Our vaccine candidate, SchistoShield, is based on an approximately 87 kDa large subunit of calcium activated neutral protease - termed Sm-p80 - combined with a potent TLR4 agonist-based adjuvant. SchistoShield has been shown to prevent disease throughout the parasitic life cycle - including egg, juvenile, and adult worm stages - in numerous animal models up to and including baboons.

Limb-girdle muscular dystrophy type 2A (LGMD R1 Calpain 3-Related, LGMD2A/R1), an autosomal recessive disorder, is characterized by progressive muscle weakness with a prominent presentation in the proximal limb girdle muscles. LGMD2A/R1, which is caused by variants in calcium-activated neutral proteinase 3 (), is the most common. The present study aimed at identifying the clinically significant variants in a Chinese family with LGMD2A/R1 and exploring the genotype-phenotype correlations.

The aim of this study was to associate single nucleotide polymorphisms () of the bovine calcium-activated neutral protease µ-calpain, calpastatin, diacylglycerol--acyltransferase, adipose fatty acid binding protein, retinoic acid receptor-related orphan receptor C (), and thyroglobulin () gene with intramuscular fat content (). Non-small cell lung cancer (NSCLC) accounts for a high proportion of lung cancer cases globally, but early detection remains challenging, and insufficient oxygen supply at tumor sites leads to suboptimal treatment outcomes. Therefore, the development of core-shell Au@Pt-Se nanoprobes (Au@Pt-Se NPs) with peptide chains linked through Pt-Se bonds was designed and synthesized for NSCLC biomarker protein calcium-activated neutral protease 2 (CAPN2) and photothermal therapy (PTT) enhancement. The NP can be specifically cleaved by CAPN2, resulting in fluorescence recovery to realize the detection.
