The universal chromophore of visual pigments in higher animals is 11-cis retinaldehyde. The final step in the biosynthetic pathway generating this compound is catalyzed by 11-cis retinol dehydrogenase, a membrane-bound enzyme abundantly expressed in the retinal pigment epithelium of the eye. In this work we demonstrate that the primary structure of human 11-cis retinol dehydrogenase is highly conserved with 91% identity to the bovine enzyme. The gene encoding 11-cis retinol dehydrogenase spans over approximately 4.1 kb of DNA and is divided into four translated exons. Analysis of a panel of somatic cells hybrids and fluorescence in situ hybridization on metaphase chromosomes revealed that the gene is located on chromosome 12q13-q14. Due to the unique role of 11-cis retinol dehydrogenase in the generation of visual pigments, it is a candidate gene for involvement in hereditary eye disease.

At the maternal-fetal interface, tightly regulated levels of retinoic acid (RA), the physiologically active metabolite of vitamin A, are required for embryo implantation and pregnancy success. Herein, we utilize mouse models, primary human cells, and pharmacological tools to demonstrate how depletion of RA signaling via RA receptor (RAR) disrupts implantation and progression of early pregnancy. To inhibit RAR signaling during early pregnancy, BMS493, an inverse pan-RAR agonist that prevents RA-induced differentiation, was administered to pregnant mice during the peri-implantation period.

Though nanozymes are becoming promising alternatives to natural enzymes due to their superior properties, constructing nanozyme with high specificity is still a great challenge. Herein, with Cu2+ as an active site and adenine as a ligand, Adenine-Cu-PO4 is synthesized in phosphate-buffered saline. As an oxidase mimic, Adenine-Cu-PO4 could selectively catalyze oxidation of ascorbic acid (AA) to dehydroascorbic acid, but not universal substrates (3,3',5,5'-tetramethylbenzidine (TMB), 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) and 2,4-dichlorophenol (2,4-DP)), small biomolecules (dopamine, glutathione, glucose, galactose), other vitamins (vitamin A acid, vitamin B1, vitamin K1) and even dithiothreitol (a common interference of AA).
