3-Aminobenzamide is an inhibitor of poly-(ADP-ribosyl)ation. In concentrations from 3 to 10 mM it reduced the collagenase activity in culture supernatants of interleukin-1 beta-stimulated rabbit synovial fibroblasts. 3-Aminobenzoate, not an inhibiter of poly(ADP-ribosyl)ation, had no effect on collagenase activity at a concentration of 10 mM. We concluded that poly(ADP-ribosyl)ation plays a role in the induction of the expression of collagenase and that 3-aminobenzamide can inhibit this process.

Interleukin-1 (IL-1) plays a crucial role in the progression of OA, and its receptor antagonist (IL-1Ra), a natural IL-1 inhibitor, represents a promising therapeutic target by obstructing the IL-1 signaling pathway. In anterior cruciate ligament (ACL) repair methods, the continuous enzymatic erosion of synovial fluid can impede healing and potentially lead to repair failure, as well as exacerbate articular cartilage wear, resulting in joint degeneration. Inspired by the blood clot during medial collateral ligament healing, we developed a composite scaffold comprising collagen (1 %, w/v) and polyvinyl alcohol (5 %, w/v) combined with platelet-rich plasma (PRP). The composite scaffold provides a protective barrier against synovial erosion for the ruptured ACL, while simultaneously facilitating tissue repair, thereby enhancing the efficacy of ACL repair techniques.
